异源三倍体为父本的百合杂交后代倍性调查及FISH分析 被引量:5

摘要 本研究通过调查异源三倍体为父本的百合杂交后代倍性及其追踪染色体来源,来探讨异源三倍体的遗传规律。采用核型分析方法,对以雄性不育的二倍体亚洲百合GHC-01为母本,以Longiflorum×Asiatic系列的异源三倍体百合‘Ceb Dazzle’为父本杂交获得的30个杂交后代及其父母本进行了染色体数目的统计,同时利用45S rDNA荧光原位杂交技术,对父母本以及4个杂交后代进行了45S rDNA荧光原位杂交分析。核型分析结果表明,GHC-01和‘Ceb Dazzle’的染色体数目分别为24(2n=2x=24)和36(2n=3x=36),30个杂交后代中染色体数目为24、25、26、35、36的分别有13、10、4、2、1个基因型的杂交后代;荧光原位杂交结果表明,GHC-01和‘Ceb Dazzle’分别有11个和14个45S rDNA荧光原位杂交位点,4个所研究的杂交后代中均有不同数目的典型父本染色体。基于倍性统计和荧光原位杂交结果,对杂交后代的早期鉴定以及异源三倍体在育种中的作用进行了讨论。 In order to explore the genetic rule of allotriploid, we studied the ploidy level and the origin of the pro- genies' chromosome from cross using allotriploid Lilium as male. The chromosome number of the parents and the thirty progenies from cross between the male-sterile Asiatic hybrid Lilium GHC-01 ( ♀ ) and the Longiflortmax Asiatic hybrid Lilium 'Ceb Dazzle' ( ♂ ) were studied by karyotype analysis. Meanwhile, four progenies and their parents were also analyzed by 45S rDNA fluorescence in situ hybridization (FISH). The karyotype analysis of result indicated that the chromosome number of GHC-01 and 'Ceb Dazzle' was 24 (2n=2x=24) and 36 (2n=3x= 36) respectively. The progeny number of which had 24, 25, 26, 35and 36 chromosomes were 13, 10, 4, 2 and 1 respectively in all the thirty progenies. The fluorescence in situ hybridization of result showed that the number of the 45S rDNA sites about GHC-01 and 'Ceb Dazzle'were 11 and 14 respectively. There were different numbers of typical chromosome contributed by male in the four progenies. Based on the result of the ploidy level statistics and FISH, the early identification of hybrid and the perspective of allotriploid as male in the polyploidy breeding oro^am of Lilium were discussed.
