摘要
目的在前期采用microPET观察到超早期针刺对脑缺血葡萄糖代谢改善的基础上,从能量代谢角度进一步探讨针刺抗脑缺血损伤的作用机理。方法采用微创开颅电凝法制作大鼠急性脑缺血模型,随机数字表法分为4组:正常组、模型组、穴位组、非穴位组,每组10只。穴位针刺取百会、水沟针刺(频率120次/min,捻转泻法1 min,每次5 min,留针30 min)。非穴位针刺分别在百会、水沟左侧旁开5 mm处进针,避开穴位,手法同穴位针刺。采用免疫组织化学法检测缺血侧皮质区域GLUT1、GLUT3的表达。结果造模成功后,缺血侧皮质区域GLUT1、GLUT3含量在大鼠脑缺血后明显下降,与正常组比较,有极显著性差异(P<0.01);穴位组大鼠GLUT1、GLUT3含量较模型组及非穴位组比较明显升高(P<0.05);而非穴位组与模型组水平接近(P>0.05)。结论穴位针刺可以有效调节脑缺血大鼠缺血侧皮质区域GLUT1、GLUT3的表达,改善能量代谢,发挥抗脑缺血损伤的作用,进一步为临床针刺抗脑缺血提供实验依据。
Objective: To study on the mechanism of anti cerebral ischemia acupuncture from the early micro- PET observation and the very early stage of cerebral ischemia acupuncture which improved glucose metabolism. Methods: The model of acute cerebral ischemia in rats made by minimally invasive craniotomy electrocoagula- tionthe were randomly divided into 4 groups such as the normal group,the model group,the acupuncture group, the non-acupoint group with 10 rats in each group. Acupoint acupuncture involved Baihui (DU20) and Shuigou (DU26) acupuncture (frequency at 120/min,twirling reinforcing-reducing for lmin and retaining the needle for 30min). Non-acupoint acupuncture needle were Baihui (DU20) and Shuigou (DU26) respectively in its left side about 5 mm,avoiding the acupoints. The needle way was the same with acupoint acupuncture. Expressions in- cluding GLUT1 and GLUT3 were detected by immunohistochemistry in ischemic cortex. Results: After the suc- cess of modeling,the count of ischemic cortex area GLUT1 and GLUT3 were decreased obviously in rats after cerebral ischemia, compared with the normal group ,there is a significant difference between 2 groups (P〈 0.01 ). The content of GLUT1 and GLUT3 in acupoint group rats compared with model group and non-acupoint group were increased obviously (P〈 0.05). But non-acupoint group compared with model group, there was no significant difference in the above index (P 〉 0.05). Conclusion: Acupuncture can improve the ischemic rat cerebral ischemia cortex area energy metabolism, act against cerebral ischemia injury, and further for clinical acupuncture in treating cerebral ischemia provide experimental basis.
