摘要
以ACC(1-氨基环丙烷-1-羧酸)为唯一氮源,采用PAF、DF 和ADF培养基定向富集的方法筛选具有ACC脱氨酶耐盐活性的促生菌株,并确定其产酶条件。结果表明:筛选出53株活性菌株,其中jz1和jz5的活性较高,分别为0.2802U·mg^-1和0.2415U·mg^-1;其产酶条件的最适pH 值为7.5,最佳培养时间为24-42h,最适温度为25-30益,随着底物浓度的增加酶活性上升。筛选出的活性菌株对黄瓜种子均有不同程度的促生作用,其中jz1 和jz6 处理较为明显,处理后黄瓜根长103.19mm,下胚轴长52.89mm。
With 1-aminocyclopropane-1-carboxylic acid (ACC) as the only source of nitrogen, the methods of directional enrichment of PAF, DF and ADF medium were applied to screen the salt resistant growth-promoting strains of ACC deaminase, and determine the enzyme producing conditions. Fifty three strains were screened as active strains in which jzl and jz5 were the most activated,(0.2802±0.0033)U/mg and (0.2415±0.0058), respectively. The screened active strains had different growth-promoting effects on cucumber seeds. Jzl and jz6 were more active than others, with the root length 103.19±0.17mm and 52.89+0.06mm, respectively. The optimum pH and temperature of the enzyme producing conditions were 7.5 and 25-30℃ respectively, and the best incubation time was 24-42h. With the increasing of the substrate concentration, the enzyme activity would rise.
