密骨胶囊对自然衰老雄性大鼠骨髓Smadl、Smad5、Smad6基因表达的影响

摘要 目的研究密骨胶囊对自然衰老雄性大鼠股骨骨髓中Smadl、Smad5、Smad6基因表达的影响。方法取12月龄自然衰老SD雄性大鼠100只随机分为2组,每组50只。自鼠龄12月龄始,分别给予生理盐水、密骨胶囊灌胃,在鼠龄14、16、18、20、22月龄时,每组各取10只大鼠,取右股骨,DEXA检测BMD,取左股骨骨髓,RT—PCR半定量检测Smadl、Smad5、Smad6基因表达。结果在14~22月龄阶段,密骨胶囊组与空白对照组相比较,密骨胶囊可维持股骨BMD(P=0.797),上调Smadl、Smad5mRNA表达水平(P〈0.01),下调Smad6mRNA表达水平(P〈O.01)。结论密骨胶囊可以上调股骨骨髓中Smadl、Smad5基因表达水平,下调Smad6基因表达水平,这可能是其促骨形成、修复骨损伤作用的分子机制之一。 Objective To investigate the mechanism of Mi Gu Capsule regulating the Smadl, Smad5, Smad6 gene ex- pression of bone marrows in aging male Spraque-Dawley rats. Methods One hundred male rats of 12-month-old were ran- domly divided into 2 groups. The rats fed with Mi Gu Capsule (MG) were as therapeutic group and with normal saline as control group. Ten rats of each group were sacrificed every two months. The right femora were obtained to detect BMD by DEXA. The bone marrows of/eft femora were collected to detect the Smadl, Smad5, Smad6 gene expressions by semi RT-PCR. Result Between 14 to 22 months follow-up, the MG could maintain the femoral BMD (P=0.797), significantly improve Smadl, 5 gene expression (P〈0.01) and decrease Smad6 gene expression (P〈0.01) comparing with that of normal saline. Conclusion MG improves the bone formation by upregulating gene expression of Smadl, Smad5 and downrezulatin~, expression of Smad6 in femoral marrows.
