摘要 目的:利用组织芯片技术探讨肺癌组织中Raf激酶抑制蛋白(raf kinase inhibitor protein,RKIP)和磷酸化细胞外信号调节激酶(phosphorylation of extracelluar signal-regulated kinase,p-ERK)的表达及其与临床病理特征的关系。方法:应用免疫组织化学方法检测96例肺癌及相应癌旁正常肺组织中RKIP与p-ERK蛋白的表达情况,并分析其与肺癌临床病理学特征的关系。结果:肺癌组织中RKIP的表达低于癌旁正常肺组织,而p-ERK的表达高于癌旁正常肺组织,差异有统计学意义。其表达水平与肺癌的分化程度、TNM分期、淋巴结转移及术后生存时间有明显关系;RKIP蛋白的表达水平与p-ERK呈负相关(P<0.05)。结论:RKIP低表达与肺癌的侵袭、转移密切相关,RKIP可能是肺癌的一个转移抑制蛋白,RKIP表达下调导致p-ERK的过表达从而促进肺癌细胞的侵袭和转移。 Objective To investigate the expression of raf kinase inhibitor protein (RKIP) and phospho- rylation of extracelluar signal-regulated kinase (p-ERK) in lung carcinoma by tissue microarray technique, and to explore its correlation with the clinicopathologic features. Methods RKIP and p-ERK protein expression were detected by using immunohistochemistry in 96 lung carcinoma specimens and 96 non-tumor lung samples. Relationship between RKIP and p-ERK expression and clinicopathological parameters was also analyzed. Results Immunohistochemistry assay revealed that RKIP level was lower in tumor tissues than that in normal lung tissues. But p-ERK positive staining rate was significantly higher in tumor tissues than that in normal lung tissues. The difference was significant (P 〈 0.05). RKIP and p-ERK expression was correlated with the different differentiation of tumor, TNM staging, the development of lymph node metastasis and the survival time (P 〈 0.05, respectively). RKIP expression was negatively correlated with ERK activation in lung carcinoma (P 〈 0.05). Conclusion Expression of RKIP and p-ERK was significantly correlated with invasion and metastasis of lung carcinoma. RKIP may be a lung carcinoma metastasis suppressor, and the decreased RKIP expression was associated with the increased metastasis capability of lung carcinoma cells through activating ERK.
