吡格列酮联合瑞舒伐他汀有效抑制动脉粥样硬化的作用 被引量:3

摘要 目的观察吡格列酮和瑞舒伐他汀单独或联合应用对高脂血症兔主动脉斑块面积和三磷酸腺苷结合盒转运体A1(ABCA1)的影响,探讨抑制动脉粥样硬化(AS)的机制。方法 50只新西兰白兔随机分为5组,每组10只:正常对照组,正常饮食;高脂肪饮食组,单纯饲喂高脂肪饲料;吡格列酮组,在单纯高脂肪饲料的基础上,给予灌喂吡格列酮5 mg/(kg·d);瑞舒伐他汀组,在单纯高脂肪饲料的基础上,给予灌喂瑞舒伐他汀1.5 mg/(kg·d);吡格列酮+瑞舒伐他汀组,在单纯高脂肪饲料的基础上,给予灌喂吡格列酮5 mg/(kg·d)、瑞舒伐他汀1.5 mg/(kg·d)。实验后20周分别利用流式细胞术检测的ABCAl表达量,利用酶法测定兔的血脂,并利用专业图像分析软件分析兔主AS面积。结果与正常对照组相比,吡格列酮组与吡格列酮+瑞舒伐他汀组均提高血浆高密度脂蛋白胆固醇、载脂蛋白A1-Ⅰ水平,并显著增加巨噬细胞的ABCA1表达量。同时使主AS面积明显减少。其中吡格列酮+瑞舒伐他汀组的变化较吡格列酮组、瑞舒伐他汀组更加明显。结论瑞舒伐他汀和吡格列酮可能通过抑制外周血单核细胞ABCA1的表达发挥抗AS作用,且两者联合应用效果更佳。 Objective To investigate the effects of pioditazone and rosuvastatin mono-therapy or combination therapy on aortic atherosclerotic area and ATP-binding cassette transporters A1 (ABCA1) synthesis in atherosclerosis (AS) rabbits. To explore the anti- atherosclerosis mechanism of pioditazone. Methods Fifty rabbits were randomly divided into four groups (10 each) : control group, high cholesterol diet group(only high cholesterol diet for 20 weeks),pioditazone group[the same high cholesterol diet plus pioditazone 5 mg/(kg.d) - for 20 weeks]. Rosuvastatin group [the same high cholesterol diet plus Rosuvastatin 1.5 mg/(kg. d) - for 20 weeks]. rosuvastatin combined with pioditazone group [the same high cholesterol diet plus pioditazone 5 mg/(kg .d) combined with Rosuvastatin 1.5 mg/(kg.d) for 20 weeks]. After 20 weeks ABCA1 expression were evaluated by flow cytometry. Enzymatic methods were used to test serum lipids levels,and area of AS in aorta was calculated by professional image analysis software. Results Compared with control group,pioditazone group and rosuvastatin combined with pioditazone group showed high-density lipoprotein cholesterol (HDL-C) and apolipoprotein A1 (apoA1) levels were increased,ABCA1 expressions in the cells were higher,and AS area in aorta were decreased.Accordingly,the changes in rosuvastatin combined with pioditazone group were more significantly than in pioditazone group and rosuvastatin group. Conclusion Rosuvastatin combined with pioditazone attenuate atherosclerosis through up-regulating ABCA1 secretion in peripheral blood monocytes. Synergistic administration of pioditazone and rosuvastatin exertsed more significant effects of anti-atherosclerosis.
