针刺、神经生长因子对脑瘫幼鼠智力及脑海马细胞凋亡基因的影响 被引量:7

摘要 目的:观察针刺和神经生长因子(NGF)对脑瘫模型幼鼠智力恢复以及脑海马组织BCL-2、BAX、Casp3基因mRNA表达的影响。方法:采用缺血缺氧(HIBD)的方法制备幼鼠脑瘫的模型,幼鼠出生后7d称重、实验分组、造模。实验分为5组:正常组、假手术组、模型组、单纯针刺组、针刺+NGF组。术后3~15d按计划给予相应干预。术后18d取材,用荧光定量PCR(SYBRGreenI染料法)检测其脑海马组织BCL-2、BAX、Casp3基因mRNA表达:用TUNEL法检测其脑皮质神经细胞凋亡情况。结果:与模型组相比,单纯针刺组和针刺+NGF组幼鼠的脑细胞抗凋亡BCL-2基因mRNA表达A值均数更高;BAX、Casp3基因mRNA表达A值均数更低:脑皮质组织细胞凋亡指数均数更低,且针刺+NGF组凋亡指数比单纯针刺组更低。结论:在抗脑神经细胞凋亡方面.针刺对HIBD幼鼠都有明显的治疗作用,加用NGF后效果更好。 Objective To observe the effects of acupuncture and nerve growth factor (NGF) on intelligence recovery and BCL-2,BAX,Casp3 gene mRNA of hippocampus tissues of rats model with cerebral palsy (CP). Methods Using hypoxic ischemic method to make the model of newborn rats with CP. All the rats 7 days ",ffter born were weighed, modeled and grouped, divided into five groups: normal control group, sham operated control group, model control group (hypoxic ischemic brain damage group), acupuncture group, acupuncture + NGF group. Related intervention was given on scheme 3-15 days after operation, drawn at 18th day, expression of brain nerve cell BCL-2, BAX, Casp3 mRNA were detected by fluorescence quantitative PCR (SYBRGreen I) ; nerve cell apoptosis were detected by TUNEL. Results Compared with model control group, mRNA expression of BCL-2 in acupuncture group and acupuncture + NGF group were higher; mRNA expression of BAX, Casp3 were lower; the apoptosis index of acupuncture group and acupuncture + NGF group were lower, and acupuncture group was lower than and acupuncture + NGF group. Conclusion Acupuncture is valid in anti-apoptosis, acupuncture adding NGF is better than acupuncture only.
