血管内皮抑素对三阴性乳腺癌化疗增敏的作用研究

摘要 【目的】探讨血管内皮抑素(Endostatin,ES)对三阴性乳腺癌(TNBC)化疗增敏的作用。【方法】37℃务件下,体外培养TNBC的MDA—M13-231细胞,用不同浓度多烯紫杉醇单药和重组人ES(rhES)两药混合分别孵育TNBC细胞24h、48h,用MTT法检测细胞生长抑制率。【结果】多烯紫杉醇单药不同浓度(0.1μg/mL、1μg/mL、10μg/mL、100μg/mL和1000μg/mL、)对MDA—MB-231细胞24h半数抑制浓度(IC50)分别为:-8.6、-9.4、-6.36、1.6、-7。联合使用rh-ES(0.05μg/mL恩度)和多烯紫杉醇不同浓度(0.1μg/mL、1μg/mL、10μg/mL、100μg/mL,1000μg/mL多西紫杉醇)对MDA—MB-231细胞24hIC50分别为:1.23、1.29、1.26、-0.22和0.01。【结论】提高浓度并不能增加多烯紫杉醇对MDA-MB-231细胞的抑制率。rh—ES可以增加低浓度多烯紫杉醇对TNBC的MDA—MB-231细胞的敏感性。 [Objective] To explore the effect of endostatin(ES) on chemotherapeutic sensitization of triplenegative breast cancer(TNBC). [Methods] TNBC MDA-MB-231 cells were cultured at 37℃, and incubated with different concentration of only docetaxel or mixed with recombinant human ES for 24h and 48h. MTT assay was used to detect the inhibition rate of cell growth. [Results] The inhibition rate of different concentration of docetaxel(0.1μg/mL, 1μg/mL, 10μg/mL, 100μg/mL and 1000μg/mL) on MDA-MB-231 cells were -8.6, -9.4, -6.36, 1.6 and -7, respectively. The inhibition rate of ES(0.05μg/mL) combined with different concentration of docetaxel(0.1μg/mL, μ1g/mL, 10μg/mL, 100μg/mL and 1000μg/mL) on MDA-MB- 231 cells were 1.23, 1.29, 1.26, -0.22 and 0.01, respectively. [Conclusion] Elevating the concentration can not increase the inhibition rate of docetaxel on MDA-MB-231 cells. Endostatin can increase the sensitivity of low concentration of docetaxel to TNBC MDA-MB-231 cells.
