摘要
对海洋胶原低聚肽的血管舒张和降胆固醇作用进行了研究。通过测定海洋胶原低聚肽作用下人脐静脉内皮细胞(HUVEC)的一氧化氮(NO)分泌量,对海洋胶原低聚肽的血管舒张活性进行了考察。结果显示,各实验组细胞在海洋胶原低聚肽的作用下,NO分泌量均有所增加,并且呈明显的量效关系。海洋胶原低聚肽浓度为12 g/L时,NO含量增加最多(P<0.01)。不同浓度海洋胶原低聚肽作用2h后,NO分泌量均比作用1h有所降低。通过观察人肝癌细胞株HepG2中低密度脂蛋白受体(LDLR)和3-羟基-3-甲基戊二酸单酰辅酶A还原酶(HMGCR)表达的变化,对海洋胶原低聚肽的降胆固醇作用进行了研究。结果显示,细胞培养24 h后,除2 g/L海洋胶原低聚肽组外,LDLR的表达量均有上调(P<0.05或P<0.01),其中4g/L海洋胶原低聚肽组的LDLR mRNA表达增加最多,呈极显著性差异(P<0.01);各实验组HMGCR的表达量均有下调,并有统计学差异(P<0.05或P<0.01),其中6 g/L海洋胶原低聚肽组的HMGCR mRNA表达最低。海洋胶原低聚肽具有促进血管舒张、降低胆固醇的作用。
The vasorelaxation and cholesterol reducing activities of marine collagen oligopeptides (MCOP) were investigated. The vasorelaxation activity of MCOP was evaluated through determination of the secretion of nitric oxide (NO) in human umbilical vein endothelial cell (HUVEC). The results showed that MCOP could increase the secre- tion of NO in all the test groups, and the vasorelaxation activity of MCOP exhibited a dose-dependent relationship. At the concentration of 12g/L, the effect was the most significant (P 〈0.01 ). The secretion of NO 1 h after addition of MCOP was higher than that 2 h after addition. The cholesterol reducing activities of MCOP was evaluated by observing the influence of MCOP on expression of low density lipoprotein receptor (LDLR) and 3-hydroxy-3-methylglutaryl co- enzyme A reductase (HMGCR) genes. The results showed that after treatment with MCOP for 24h, LDLR mRNA lev- els were obviously increased in all test groups (P 〈 0.05, P 〈 0.01 ) except for 2g/L MCOP group, with LDLR mR- NA level in 4 g/L of MCOP group increased the most (P 〈 0.01 ). HMGCR mRNA levels were all decreased in all test groups (P 〈 0.05, P 〈 0.01 ) , with HMGCR mRNA level in 6g/L of MCOP group decreased the most. The re- suits in this research indicated that MCOP exhibited vasorelaxation and cholesterol reducing activities.
