摘要
目的观察厚朴提取物对大鼠CYP2D6亚型酶的影响。方法将16只成年雄性Wistar大鼠随机分为对照组和实验组,2组分别经口给予生理盐水和厚朴提取物1周,采用高效液相色谱法(HPLC)测定大鼠尿样及肝微粒体中CYP2D6的探针药物右美沙芬(DM)的代谢率,观察厚朴提取物对CYP2D6亚型酶活性的影响,并通过特异性抑制剂确定肝微粒体重组系统中厚朴提取物对CYP2D6亚型酶活性的影响。结果实验组大鼠尿样及体外肝微粒体中DM代谢率均明显低于对照组,差异有统计学意义;厚朴提取物组肝微粒体中DM代谢率显著低于西咪替丁组和对照组,差异有统计学意义,而西咪替丁组和对照组比较,差异无统计学意义。结论厚朴提取物可有效抑制CYP2D6亚型酶的活性,且抑制能力优于西咪替丁。
Objective To observe the influence of magnolia bark extract on the activity of CYP2D6 subtype enzyme. Methods Sixteen adult Wistar rats were randomly divided into control group and experimental group. The rats in two groups were orally given normal saline and magnolia bark extract for 1 week. High performance liquid chromatography (HPLC) was used to measure the metabolic rate of dextromethorphan (DM) in the urine and liver micrommal reconstituted sys- tem of rats to observe the influence of magnolia bark extract on the activity of CYP2D6 subtype en- zyme, and its influence on the activity of CYP2D6 subtype enzyme was also observed in the liver mi- crosomal reconstituted system by specific inhibitors. Results DM metabolic rates in urine and liver micromme of rats in the experimental group were distinctly lower than in the control group, and the difference was statistically significant. DM metabolic rate in the liver microsome of rats in magnolia bark extract group was markedly lower than in cimetidine group and the control group, and the dif- ference had statistical significance, whereas there was no statistical significance between cimetidine group and the control group. Conclusion Magnolia bark extract can effectively inhibit the activity of CYP2D6 subtype enzyme, and its inhibitory capability is superior to cimetidine.
