不同浓度的胰岛素对SW480肿瘤细胞分泌IGF-1的影响

摘要 目的探讨不同浓度的胰岛素对SW480肿瘤细胞IGF-1表达的影响。方法体外常规培养SW480肿瘤细胞。根据所用培养液外加胰岛素浓度的不同,将实验细胞分为七组:0组(对照组)RPMI 1640培养液;第1组RPMI 1640培养液+5μIU/ml胰岛素;第2组RPMI1640培养液+25μIU/ml胰岛素;第3组RPMI 1640培养液+50μIU/ml胰岛素;第4组RPMI 1640培养液+75μIU/ml胰岛素;第5组RPMI 1640培养液+100μIU/ml胰岛素;第6组RPMI 1640培养液+125μIU/ml胰岛素。每组设3个复孔,置37℃、5%CO_2饱和湿度的培养箱培养48 h,培养结束后收集细胞培养上清,ELISA法检测IGF-1的浓度。结果高浓度的胰岛素组(第5组、第6组,培养液中分别含100、125μIU/ml的胰岛素)IGF-1的表达(286.1 8±23.98、296.28±32.57)高于无胰岛素的对照组(193.22±22.77),差异有统计学意义(P<0.01);而1、2、3、4组(培养液中分别含5μIU/ml、25μIU/ml、50μIU/ml、75μIU/ml的胰岛素)IGF-1的表达((192.19±25.98、201.10±20.42、204.88±20.40、207.05±-30.44)与无胰岛素的对照组(193.22±22.77)比较,差异无统计学意义(P>0.05)结论高浓度的胰岛素增加了SW480肿瘤细胞IGF-1的分泌。 Objective To observe the effects of different concentrations of insulin on the secretion of insulin-like growth factor-1 ( IGF-1 ) in SW480 tumor cells. Methods SW480 tumor cells were cultured in vitro. According to the concentrations of insulin in culture media, cells were divided into seven groups, i. e. 0 group (control group) with RPMI 1640, first group with RPMI 1640 ± insulin at 5 pJU/mL, second group with RPMI 1640 ± insulin at 25μIU/mL, third group with RPMI 1640 ±50 μIU/mL insulin, fourth group with RPMI 1640 ± insulin at 75 p, IU/mL, fifth group with RPMI 1640 ± insulin at 100 txIU/mL and sixth group with RPMI 1640 ± insulin at 125μIU/mL. After added to different culture media, cells were cultured for 48 hours, and supernatant was collected. In all the experiments, IGF-1 concentration in the supernatant was detected by specific enzyme-linked immunosorbent as- say (ELISA). Results In the high concentration of insulin groups (the fifth group and the sixth group) , the level of IGF-I was significantly higher than that in the 0 group (286.18 ±23.98 and 296.28 ±32.57 vs 193.22 ± 22.77, P 〈0.01 ). In the first, second, third, and fourth groups, compared with the 0 group, there were no statistical differences in the secretion of IGF-1 in SW480 tumor cells ( 192.19 ~ 25.98, 201.10 ~ 20.42, 204.88 ± 20.40 and 207.05 ± 30.44 vs 193.22 ± 22.77, P 〉 0.05 ). Conclusion High concentration of insulin may stimulate the secretion of IGF-1 in SW480 tumor cells.
