摘要
目的研究TBK1(TANK binding kinase 1)在胰腺癌组织中的表达,并探讨其与胰腺癌恶性生物学行为及上皮间质转化(EMT)的关系。方法应用免疫组织化学技术SP法,检测50例胰腺癌组织中TBK1、间质源性标志物vimentin蛋白的表达,分析其与患者的临床病理资料的关系,以及二者的相关性。选择20例正常胰腺组织为对照组。结果在50例胰腺癌组织中TBK1染色阳性率为70%,与对照组(20%)比较差异有统计学意义(P<0.05)。TBK1的表达与与胰腺癌的分化程度、临床分期、淋巴结转移、远处转移存在相关性,差异具有统计学意义(P<0.05);vimentin表达阳性率为42%,对照组阳性率为0,两者间差异具有统计学意义(P<0.05);vimentin的阳性表达与胰腺癌组织的淋巴转移、远处转移明显相关(P<0.05);在胰腺癌组织中TBK1的表达与vimentin表达间具有正相关性(r=0.427,P<0.05)。结论 TBK1是反映胰腺癌恶性程度的肿瘤标记物,与胰腺癌侵袭、转移密切相关,可能源于TBK1参与了胰腺癌的EMT现象。
Objective To investigate the expression of TANK binding kinase 1 ( TBK1 ) in pancreatic cancer tissues,and to explore its relations with the malignant features and epithelial-mesenchymal transition(EMT). Methods The expression of TBK1 and vimentin proteins was detected using immunohistochemistry in 50 cases of pancreatic cancer tissues and 20 cases of normal tissues, its relations with the clinicopathological features were analyzed. Results The positive expression rate was 70% (35/50) for TBK1 and 42% (21/ 50) for vimentin protein in pancreatic cancer tissues, and they were statistically higher than those in controls( P 〈 0.05 ). The positive expression rate of TBK1 was significantly associated with the degree of cell differentiation, clinical stage, lymph node metastases, and distant metastases (P 〈 0.05 ). The expression of vimentin in pancreatic cancer tissue was significantly associated with lymph node me-tastases and distant metastases(P 〈 0.05 ). There was a significantly positive correlation between the expression of TBK1 and vimentin in pancreatic cancer tissues (r = 0. 427,P 〈 0. 05). Conclusion TBK1 is a marker to refl ect the severity of pancreatic cancer, and is related to the invasion and metastasis process, which may be related to the involvement of the epithelial-mesenchymal transition.
