结直肠腺瘤及结直肠癌中TLR7 TLR9及MyD88的表达 被引量:2

摘要 目的:通过检测不同组织类型的腺瘤中Toll样受体7、9与MyD88的表达情况,探讨其在结直肠癌发生发展中的临床意义。方法:采用免疫组织化学EnVision法,检测50例增生性息肉、62例管状腺瘤、46例绒毛状腺瘤、26例混合型腺瘤、42例结直肠癌中TLR7、TLR9及MyD88的表达情况,其中8例绒毛状腺瘤与5例混合型腺瘤局灶发生癌变。结果:TLR7、TLR9的表达随着结直肠黏膜上皮病变程度增加,其阳性表达率均先升高后降低(P<0.05)。TLR7、TLR9蛋白表达均与腺瘤的病理类型及异型性相关(P<0.05),而与年龄、性别及部位无关(P>0.05)。MyD88的表达随着腺上皮病变程度增加,其阳性表达率逐级升高(P<0.05)。MyD88蛋白表达仅与腺瘤的异型性相关(P<0.05),而与年龄、性别、部位及病理类型均无关(P>0.05)。结直肠腺瘤组织TLR7、TLR9两者的表达均与MyD88表达呈负相关(P<0.05)。结论:TLR7、TLR9两因子的表达与MyD88蛋白的表达在结直肠肿瘤的发生发展中可能具有拮抗抑制作用,其联合检测能为结直肠肿瘤的早期临床诊断和治疗提供有利的生物学信息。 Objective:To investigate the role of the expression of TLR7,TLR9and MyD88in colorectal adenomas and colorectal carcinoma.Methods:The expression of TLR7,TLR9and MyD88were detected by immunohistochemistry in 50cases hyperplastic polyps,62cases tubular adenoma,46cases villous adenoma and 26cases tubulovillous adenoma,42cases colorectal carcinoma,the 8cases of villous adenoma and 5cases of tubulovillous adenoma occur carcinogenesis.Results:Expression of TLR7and TLR9with colorectal epithelial lesions degree increased,the positive rate was increased first and then decreased(P〈0.05).There were statistically differences between TLR7or TLR9expression in pathologic type and histological atypia(P〈0.05).The high expression of MyD88expression was significantly associated with histological atypia(P〈0.05).Negative correlation was found between the expression of TLR7or TLR9and MyD88in colorectal adenomas.Conclusion:The expression of TLR7,TLR9and MyD88may be mutual antagonistic relationship in colorectal carcinoma pathogenesis,which can provide a powerful biological information for the early diagnosis and treatment of colorectal cancer.
