虹鳟PDCD6基因cDNA全长的克隆与功能预测

摘要 程序性死亡因子6(PDCD6)基因编码1个含EF-hand结构域的钙离子结合蛋白。应用逆转录PCR及cDNA末端快速扩增(RACE)技术从虹鳟(Oncorhynchus mykiss)脑组织中克隆了PDCD6基因cDNA的全序列(GenBank No:FJ591154)。序列全长1 179bp,其中5’端非翻译区长48bp,3’端非翻译区长567bp,开放性阅读框长564bp,编码187个氨基酸。电子表达谱分析结果表明该基因在4种脊椎动物的多个部位或组织中均有表达。同源模建蛋白三级结构显示该蛋白具有8个主要的α螺旋结构。虹鳟PDCD6蛋白序列与斑马鱼、非洲爪蟾、人、小鼠和鸡的同源性均高于85%,表明PDCD6基因在进化过程中是高度保守的,在细胞程序性死亡过程中发挥重要作用。 Programmed cell death 6 (PDCD6) protein is an EF-hand type Ca^2+ -binding protein involved in apoptosis process. RT-PCR and RACE methods were used for the isolation of the whole cDNA of PDCD6 gene (GenBank accession number: FJ591154) from brain of rainbow trout. Sequence analysis revealed a 1 179 bp cDNA containing 48 bp 5 '-untranslated region, 567 bp 3'-untranslated region and 564 bp open reading frame encoding 187 amino acids. Electronic expression profile analysis showed that the expressions of the gene were found in several parts or tissues in four species of vertebrates. Homology modeling of tertiary structure of the protein PDCD6 showed that PDCD6 contained 8 main alpha helix. Sequence alignment between rainbow trout and zebrafish, frog, chicken, mouse and human exhibited more l han 85 identity rate of amino acid. The result indicated the PIDCD6 gene is relatively conservalive in the progress of evoIution and play an importanl role in programmed cell death Drocess.
