摘要
目的观察电针内关穴对糖尿病心肌病大鼠线粒体通透性转运孔及其机理的影响,以探讨针刺对糖尿病心肌病线粒体通透性转运孔调控各因素的相互作用。方法 30只雄性Wistar大鼠,随机分为正常组、模型组和治疗组各10只,采用一次性腹腔注射链脲佐菌素(STZ)建立糖尿病心肌病大鼠模型。治疗组电针内关穴治疗8周后,迅速取出心肌,提取线粒体,采用紫外分光光度仪检测心肌线粒体PTP开放;采用荧光光度仪,通过测试介质Rh123的荧光值检测线粒体膜电位;采用双光束紫外分光光度仪通过测试Ca2+指示剂AⅢ吸光值检测Ca2+转运。结果与模型组比较,治疗组MPTP开放检测吸光值及Ca2+转运检测吸光值明显降低(P<0.01);与模型组比较,治疗组膜电位Rh123荧光值明显提高(P<0.05)。结论糖尿病心肌病大鼠经电针内关穴治疗后MPTP高开放状态明显下降,其发生发展的机理可能与线粒体膜电位的提高与细胞内Ca2+转运的降低密切相关。
Objective: To observe the influence and the mechanism of electroacupuncturing on Neiguan(PC6) on the mitochondrial permeability transition pore of rats with diabetic cardiomyopathy. Methods 30 Wistar rats were randomly divided into the control group,the model group and the treatment group with 10 rats in each group. Af- ter quickly removing the myocardium,the mitochondrial extraction were detected by ultraviolet spectrophotometry of skeletal muscle mitochondrial PTP opening. Fluorescence photometric instrument was used to test the value of media Rh123 fluorescence detection of mitochondrial membrane potential. Dual-beam UV spectrophotometer was used to test the Ca2* indicator A HI absorbance detection of Ca2~ transport. Results: MPTP opening detecting ab- sorbance valueand Ca2~ indicator A m absorbance detection in the treatment group were more lower obviously than that in the model group(P〈 0.01 ). Rh123 fluorescence values in the treatement group were more higher sig- nificantly than those in the model group(P〈 0.05). Conclusion: MPTP high open state decrease significantly after electroacuDuncturin~ NeiCuan(PC6) points treated reduction Ca2+ translocation from intracellular.
