摘要
采用异硫氰酸苯酯(PITC)柱前衍生反相高效液相色谱法测定豆粕和发酵豆粕中17种氨基酸含量,以氨基酸C18色谱柱为分析柱,采用紫外检测器,35min内能够很好的分离17种氨基酸。氨基酸峰面积与氨基酸浓度之间的线性关系较好,加标回收率较高。试验结果表明本方法操作简单、灵敏度高,适用于豆粕和发酵豆粕中氨基酸的测定。
The 17 kinds of amino acids in soybean meal and fermented soybean meal by precolumn derivatization with phenyl isothiocyanate (PITC) with reversed-phase high performance liquid chromatography were determined. The analysis of 17 kinds of amino acids was accomplished within 35 min using C18 column as analytical column and UV detector. The linear relationship of amino acid peak area with amino acid concentration was good, and the recovery rate was high. The test results showed that this method was simple with high sensitivity, and could be applied in determination of amino acids in soybean meal and fermen- ted soybean meal
