阿霉素控释剂对大鼠C6脑胶质瘤治疗的研究 被引量:12

摘要 目的 探讨自制阿霉素控释剂在体内、外释药效果及对大鼠C6胶质瘤动物模型的治疗作用。方法 应用荧光光谱法检测阿霉素控制剂在人工脑脊液及 18只正常家兔脑内药物释放的情况 ;检测阿霉素控释剂对 10只脑内荷瘤SD大鼠生存期的影响以及不同给药方式 (局部或腹腔给药 )对 30只SD大鼠皮下接种肿瘤生长的抑制作用。结果 阿霉素控释剂在人工脑脊液和兔脑内可持续释放达 10d以上 ;局部控释化疗可明显延长脑内荷瘤大鼠的生存期 ,实验组中位生存时间为 31d。局部阿霉素控释化疗对于皮下肿瘤有明显抑制效应 ,与腹腔注射等量阿霉素化疗组相比 ,差异有非常显著性 (P <0 .0 1)。结论 控释化疗在避免系统化疗全身毒性的同时 ,增加肿瘤局部药物浓度 。 Objective To study the in vitro and in vivo control releasing effect of adriamycin following implantation of self made adriamycin containing matrix, and evaluate the local chemotherapeutic effect on rats bearing C6 gliomas. Methods The matrix with adriamycin was prepared. Gelatin and agar were used as major materials. The releasing effects of adriamycin containing matrix were determined in artificial cerebrospinal fluid or in 18 normal rabbit's brains by the method of fluorometry. The median survival time (MST) was measured in 10 SD rats bearing intracranial C6 gliomas and the inhibitory effect of regional or intraperitoneal administration of adriamycin on growth of subcutaneous tumors were evaluated in 30 rats. Results Constant release of adriamycin from the matrix (weighing 48.6 to 52.5 mg, containing adriamycin 81.2~87.2 μg) lasted over 10 days in artificial CSF or normal rabbit brains; Local controlled release of adriamycin could prolong the survival of rats bearing C6 gliomas with the median survival time being 31 days, and the growth of subcutaneous tumors was significantly inhibited. Conclusion With high regional concentration and avoidance of toxicity of systemic chemotherapy, control released chemotherapy of adriamycin remarkably inhibited the growth of C6 gliomas. [
