大鼠液压冲击脑损伤炎性细胞因子表达与神经功能评分的关系 被引量:7

The relationship between file expression of inflammatory cytokines and neuronal function score of fluid percussion brain injury in rats

摘要 目的观察大鼠液压冲击脑损伤炎性细胞因子白细胞介素一1β(IL一1β)、白细胞介素一6(IL一6)、肿瘤坏死因子一仪(TNF—Oα)、细胞问黏附分子一1(ICAM一1)表达变化,探讨其与神经功能评分的关系。方法成年雄性sD大鼠48只,制作液压冲击颅脑损伤模型,术后6h、12h、24h、3d、7d对动物进行神经功能评分,并用干湿重法、免疫组化法分别测定水肿脑组织含水量及其IL一1B、IL一6、TNF—Or.、ICAM-1表达。结果神经功能评分6h出现下降[(3.78±0.84)分],24h下降至最低值[(2.65±0.32)分],3d逐渐升高[(4.75±0.71)分];免疫组化显示水肿脑组织IL一6、IL一1β、TNF—α、ICAM一1的表达均6h开始增高,24h至3d达到高峰,7d后降低。线性相关性分析发现IL一6、IL一1β、TNF—α.、ICAM一1表达与神经功能评分呈负相关关系(相关系数分别为r:-0.912,r=-0.892,/r=一0.794,r=一0.887,P〈0.05)。结论大鼠颅脑液压冲击伤IL.1B、IL-6、TNF一仪、ICAM一1均出现高表达,与神经功能评分呈负相关,可能与炎l生因子造成的继发l生脑损伤有关。 Objective To investigate the expression of inflammatory cytokines such as interlukin-ll3 (IL- 1β ) , interlukin-6 ( IL-6 ) , tumor necrosis faetor-α ( TNF-α) and intereelluar adhesion molecule-1 ( ICAM-1 ) , and explore their relationships with neuronal fhnction score after fluid percussion brain injury in rats. Methods The experimental models were established in 48 adult rats. The water content of edematous brain and the expression of IL- 1β, IL-6, TN F-α and ICA M- 1 were ured with dry-wet measure, immunohistochemistry at 6 h, 12 h, 24 h, 3 d, 7 d after operation respectively. Compared with animals of shame operation (SO) group, neuronal func- tion score decreased in TBI grup from 6h(3280.84) ,reached to the lowest level at 24h(2.650.32) ,and grad- ually increased at 3d (4.75 0.71 ) IL-6, I1-113, TNF-tx and ICAM-limmunoreactive expression increased from 6h, reached its maxmum at 24h, fasted to 3i.h day, and began to decrease at 7d. The linear regression analysis indi- cated that expression of IL-1 , IL-6, TNF-α and ICAM-1 had negative correlations with change of neuronal function score( r=-0.912, r=-0.892 r=-0.794, r=-0887 respectively, P〈0.05).Conclusion IL-6,IL-113,TNF-a and ICAM-1 expression is upregulated following fluid percussion injury in rats, and shows negative correlations with neuronal function score.The possible mt..chanisms is that inflammatory cytokines are involved in nerve function im- pairment by inducing secondaly brain fijury.
