不同采收期薄荷中4个黄酮苷的含量测定 被引量:13

HPLC simultaneous determination of four flavone glycosides in Menthae Haplocalycis Herba during different harvest periods

摘要 目的:建立高效液相色谱同时测定薄荷中橙皮苷、香叶木苷、香蜂草苷、蒙花苷4个黄酮苷类化合物含量的方法,并测定不同采收期4个化合物的含量,观察其动态变化规律。方法:采用Sunfire C18色谱柱(4.6 mm×150 mm,5 μm),柱温30 ℃,流动相甲醇(A)-水(B),梯度洗脱(0~10 min,40%A→60%A;10~15 min,60%A→70%A),流速1 mL·min-1,检测波长285 nm。结果:橙皮苷、香叶木苷、香蜂草苷及蒙花苷4个成分均达到基线分离,其分别在0.00788~0.788、0.00884~0.884、0.0064~0.64、0.00736~0.736 μg范围内线性关系良好(r 〉0.9996);平均加样回收率(n=6)分别为100.1%、99.9%、99.8%、99.7%。橙皮苷、香蜂草苷、蒙花苷的含量在7月下旬、9月中旬较高,香叶木苷则在7月上旬、9月下旬较高。结论:含量测定方法操作简单、快速、重复性好,为薄荷药材的质量控制奠定了基础。4个黄酮苷含量随采收期不同而异,其总量最大值出现在7月上旬和9月中旬。 Objective: To establish an HPLC method for simultaneous determination of four flavone glycosides(hesperidin,diosmin,didymin and buddleoside)in Menthae Haplocalycis Herba,and to determine the content of the four components during different harvest periods to observe their dynamic changes. Methods: HPLC was performed on a Waters C18 analytical column(4.6 mm×150 mm,5.0 μm)at 30 ℃ with methanol(A)-water(B) as the mobile phase by gradient elution(0-10 min,40%A→60%A;10-15 min,60%A→70%A).The detection wavelength was 285 nm and the flow rate was 1 mL·min-1. Results: The baseline separation of these four components was achieved.The linear ranges of hesperidin,diosmin,didymin and buddleoside were 0.00788-0.788,0.00884-0.884,0.0064-0.64,0.00736-0.736 μg,respectively(r〉0.9996).The average recoveries(n=6)were 100.1%,99.9%,99.8% and 99.7%,respectively.The contents of hesperidin,didymin,buddleoside were higher in the late July and the middle of September;the content of diosmin was higher in the early July and the late September. Conclusion: This method is proved to be simple,quick,repeatable,and can be used for quality control of Menthae Haplocalycis Herba.The contents of the four flavonoid glycosides are different in different harvest time.The maximum contents appear in early July and the middle of September.
