雾化吸入伊洛前列素对脂多糖诱导大鼠不同程度肺损伤的影响

摘要 目的探讨雾化吸入伊洛前列素对脂多糖(LPS)诱导大鼠不同程度肺损伤的影响。方法雄性SD大鼠40只,按随机数字表分为五组(n=8)。C-NS组腹腔注射生理盐水,LPS1与LPS2组分别腹腔注射LPS 6、8mg/kg制备大鼠肺损伤模型,LPS1与LPS2组又分两个亚组,LPS1-NS、LPS2-NS组腹腔注射LPS后雾化吸入生理盐水,LPS1-IL、LPS2-IL组腹腔注射LPS后雾化吸入伊洛前列素2μg/kg。开胸处死大鼠取肺支气管肺泡灌洗液(BALF),测定BALF中P-selestin、IL-1β;测定BALF和血浆中蛋白含量,计算肺通透指数;行肺组织病理学评分和中性粒细胞(PMN)计数。结果各LPS组肺通透指数、P-selestin、IL-1β较C-NS组明显升高(P<0.05),但LPS2-IL组肺通透指数明显低于LPS2-NS组(P<0.05),LPS1-IL、ILPS2-IL组P-selestin明显低于LPS1-NS、LPS2-NS组(P<0.05)。LPS1-IL组和LPS2-IL组病理学评分和PMN计数均明显低于LPS1-NS组和LPS2-NS组(P<0.05)。结论伊洛前列素雾化吸入提高氧分压,减轻肺损伤早期的炎症反应,降低肺毛细血管通透性,抑制PMN在肺内聚集,减轻LPS所致肺损伤。 Objective This study is to investigate the effects of iloprost aerosol inhalation on different degree of Lipopolysacharide(LPS)-induced lung injury in rats. Methods Forty rats were randomly distributed into five groups (n =8). The rats in group C-NS was given normal saline (NS) intraperitoneally and the rats in groups LPS1 and LPS2 were given 6 mg/kg and 8 mg/kg of LPS respectively. The group LPS1 and LPSg were then divided into two subgroups: one was given NS aerosol inhalation and the other was intervened with iloprost. The arterial bold gas from blood samples, the lung permeability, and the expression of P-selestin and IL-1β from lung bronchoalveolar lavage fluid (BALF) were measured. The lung tissue was under pathological studies. Results The lung permeability in group LPS2-IL was significant lower as compared with the group LPS2-NS (P〈0.05). The concentration of IL-1β and P-selestin in BALF were increased after LPS induction. The P-selestin in the groups intervened with iloprost were significant lower when compared with the NS control group(P〈0. 05). Lung tissue pathology scores in groups intervened with iloprost were significantly lower than that in the NS control group (P〈0. 05). The PMN filtrations in groups exlmsed to iloprost were significantly lower than the NS control group (P〈0. 05 ). Conclusion Iloprost aerosol inhalation improved oxygen partial pressure, reduced lung injury in early inflammatory reaction, reduced pulmonary capillary permeability,and alleviated PMN accumulation in LPS-induced injured lung.
