两种肉桂提取物对高糖高脂诱导大鼠胰岛素抵抗的作用研究 被引量:4

摘要 目的:比较锡兰肉桂和肉桂提取物对高糖高脂诱导大鼠胰岛素抵抗的治疗作用。方法:将wistar大鼠分为4组(n=12),对照组给予普通饲料,模型组、锡兰肉桂组和肉桂组给予高糖高脂饲料,共喂养14周。第10周后锡兰肉桂组给予锡兰肉桂提取物600 mg/kg·d、肉桂组给予肉桂提取物600 mg/kg·d,模型组给予同体积蒸馏水。14周后检测空腹血糖水平(FBG)、腹腔注射葡萄糖耐量试验(IPGTT)、空腹血清胰岛素水平(FIns)、胰岛素抵抗指数(HOMA-IR)、血清肿瘤坏死因子α(TNF-α)、甘油三酯(TG)、大鼠内脏脂肪及附睾脂肪重量、肝糖原含量及肝脏葡萄糖转运体2(GLUT2)的表达水平。结果:在IPGTT中,锡兰肉桂组和肉桂组血糖曲线下面积分别为(115.2±16.1)mmol·min/L和(112.0±13.6)mmol·min/L,较模型组显著降低(P<0.01);锡兰肉桂组的FIns、HOMA-IR、TNF-α和TG分别为(17.89±3.15)μIU/mL、5.96±0.93、(1.38±0.37)μg/L和(0.67±0.25)mmol/L,肉桂组则分别为(18.02±2.56)μIU/mL、6.36±1.22、(1.45±0.31)μg/L和(0.66±0.21)mmol/L,均较模型组显著降低(P<0.01或P<0.05);锡兰肉桂组的内脏脂肪和附睾脂肪占体重百分比分别为(0.83±0.13)%和(0.55±0.16)%,肉桂组则分别为(0.79±0.15)%和(0.56±0.13)%,均较模型组显著降低(P<0.01或P<0.05);锡兰肉桂组的肝糖原含量和GLUT2蛋白表达分别为(1.66±0.27)mg/g和(49.8±5.4)mg/g protein,肉桂组则分别为(1.65±0.23)mg/g和(50.9±3.2)mg/g protein,均显著高于模型组(P<0.01)。锡兰肉桂组和肉桂组比较,各指标均无显著差异(P>0.05)。结论:锡兰肉桂和肉桂提取物均能够减轻高糖高脂诱导的大鼠胰岛素抵抗,两种肉桂提取物的作用效果无明显差别。 Objective :To compare the effects of the Ceylon cinnamon and Chinese Cassia extracts on the high sugar and high fat- induced insulin resistance in rats. Methods:Wistar rats were divided into four groups (n = 12). The control group was fed with common diet. The insulin resistant (IR) group, the Ceylon cinnamon(CLC) group, and the Chinese Cassia(CNC) group were fed with high sugar and high fat diet for 14 weeks. At the last 4 weeks, the CLC group was treated with 600 mg/( kg· d) of Ceylon cinnamon extract, and the CNC group was treated with 600mg/( kg · d) of Chi- nese Cassia extract, and the IR group was treated with distilled water. All the animals were sacrificed 14 weeks later, the blood and liver samples were collected. The body weight, the visceral fat weight/body weight(VFW/BW) and epididy- mal fat weight/body weight(EFW/BW) ratios, FBG, IPGTT, fasting serum insulin( Fins), HOMA - IR, TNF - α, TG, liver glycogen and GLUT2 were examined. Result : In glucose tolerance test, the area under the curve of time - blood glucose of CLC [ ( 115.2 ±16.1 )mmol ] min/L]and CNC [ ( 112.0 ±13.6)mmol · min/L ] groups were significantly smaller than that of the IR group(P 〈 0.0l ). In comparison to Fins, HOMA -IR, TNF-α, and TG in IR group,all of the levels were significantly lower in CLC [ ( 17.89 ±3.15 ) μIU/mL, 5.96 ± 0.93, ( 1.38 ± 0.37) μg/L, (0.67 ±0.25 ) mmol/L rgroup: and CNC [ ( 18.02±2.56) μIU/mL, 6.36 ±1.22, ( 1.45 ±0.31 ) μg/L, (0.66 ± 0.21 ) mmol/L] group(P 〈 0.01 or P〈 0.05 ). The VFW/BW and EFW/BW ratio in CLC I (0- 83 ±0.13 ) %, (0.55 ±0.16) % ] or CNC [ (0.79 ±0.15 )%, (0.56 ± 0.13 )% ] groups were significantly lower than that of the IR group( P 〈 0.01 ). The hepatic glycogen content of CLC [ ( 1.66±0.27) mg/g] and CNC [ ( 1.65±0.23) mg/g] groups were significantly high- er than that of the IR group(P 〈 0.01 ).
