摘要 目的研究实验性自身免疫性脑脊髓炎(EAE)大鼠脊髓载脂蛋白E(Apo E)的表达及其可能的来源。方法将48只雌性Wistar大鼠随机分为EAE组(32只)和正常对照组(16只)。给EAE组大鼠足垫注射豚鼠全脊髓匀浆制作EAE模型。EAE组和正常对照组大鼠脊髓进行常规HE染色,以及apo E、胶质纤维酸性蛋白(GFAP)免疫组化染色。结果 HE染色后光镜下EAE组大鼠脊髓每个节段均可见大量炎性细胞浸润,小血管明显扩张,炎性细胞聚集在血管周围形成袖套样改变,典型的病理学改变主要分布在白质以及灰白质交界区域;正常对照组大鼠脊髓无类似改变。免疫组化染色后,EAE组大鼠脊髓白质和灰白质交界区域的Apo E阳性细胞明显多于正常对照组;白质中的GFAP阳性细胞明显多于正常对照组。大多数Apo E阳性细胞为GFAP阳性。结论 EAE大鼠脊髓的Apo E表达增高,提示其参与多发性硬化的免疫反应机制和脂质转运、神经髓鞘修复机制,并与星形胶质细胞关系密切。 Objective To study the expression of apolipoprotein E ( Apo E ) of spinal cord in experimental autoimmune encephalomyelitis (EAE) rats, and its possible source. Methods Forty-eight female Wistar rats were randomly divided into EAE group ( 32 rats) and normal control group ( 16 rats). With guinea pig spinal cord homogenate to reduce EAE model. The spinal cord of the rats in EAE group and normal control group was staining through HE, and the expression of Apo E antibody and glial fibriliary acidic protein ( GFAP ) was assayed by immunohistochemical staining. Results After HE staining,Perivascular "cuffing" change, including vasodilatatiun and infiltration of inflammation cells, in each spinal cord segment was observed through HE staining under an optical microscope. The typical pathological change mainly distributed in both white matter and junction area of grey-white matter. No similar pathological change was found in normal control group. After immunohistochemical staining, Apo E immunohistostaining cells in white matter and junction area of grey-white matter of spinal cord of the rats in EAE group was markedly increased, and GFAP immunohistostaining cells was markedly increased in white matter than normal control group. Most Apo E immunohistostaining cells were also GFAP immunohistostaining. Conclusions The increase of Apo E expression,related with astrocyte closely, suggests the abnormality of immunological reaction,lipid transport and nerve myelin sheath repair mechanism in multiple sclerosis.
