付罐疗法对延胡索乙素贴剂透皮吸收的影响 被引量:5

Effect of Fu's Cupping Therapy on Transdermal Absorption of Tetrahydropalmatine Patches

摘要 目的:考察付罐疗法对延胡索乙素贴剂透皮吸收的影响。方法:以AUC0-t,T max,C max的综合评分为评价指标,通过正交试验考察罐法、压力、罐疗时间3个因素对延胡索乙素贴剂透皮吸收的影响。采用UPLC-MS/MS测定延胡索乙素贴剂的血药浓度变化,流动相乙腈-水(70∶30),流速0.3 mL·min-1,自动进样器温度30℃,柱温30℃,进样量3μL,电喷雾电离源(ESI+),质荷比(m/z)356.27,毛细管电压3.38 kV,锥孔电压38 V,离子源温度150℃,脱溶剂气温度350℃,锥孔反吹气流量50 L·h-1,脱溶剂气流量650 L·h-1,碰撞气流量0.16 mL·min-1。结果:最佳付罐疗法条件为刮罐,压力-0.02 MPa,时间15 min。结论:付罐能显著提高延胡索乙素贴剂的透皮吸收,付罐疗法可作为一种新的物理促渗方法。 Objective: To investigate effect of Fu's cupping therapy on transdermal absorption of tetrahydropalmatine patches. Method: With composite score of AUC0-t,Tmax and Cmax as index,orthogonal design was adopted to optimize technology parameters of Fu's cupping therapy by taking tank methods,pressure and time as factors.UPLC-MS/MS was used to determine plasma concentration of tetrahydropalmatine in tetrahydropalmatine patches,mobile phase of acetonitrile-water(70:30),flow rate 0.3 mL·min-1,autosampler temperature 30℃,column temperature 30℃,injection volume 3 μL,electrospray ionization(ESI+),m/z 356.27,capillary voltage 3.38 kV,cone voltage 38 V,ion source temperature 150℃,desolvation temperature 350℃,anti-cone gas flow rate 50 L·h-1,desolvation gas flow rate 650 L·h-1,collision gas flow rate 0.16 mL·min-1. Result: The best conditions of Fu's cupping therapy was:scraping tank,low pressure (-0.02 MPa) and time of 15 min. Conclusion: Fu's cupping therapy could significantly improve transdermal absorption of tetrahydropalmatine patches,which was a new physical method for promoting transdermal absorption.
