外源性一氧化碳释放分子2对同种异基因皮肤移植小鼠T淋巴细胞亚群的影响

摘要 目的:运用流式细胞技术观察经过外源性一氧化碳CORM-2处理7 d的小鼠同种异基因皮肤移植模型受体小鼠脾脏淋巴细胞亚群CD4+/CD8+的变化。方法:以清洁级ICR小鼠、BALB/C小鼠分别作为供体和受体建立异体皮肤移植模型,共分4组:1组假手术组,2组同种异基因皮肤移植组,3组同种异基因皮肤移植+CORM-2实验组,4组异体皮肤移+iCORM-2残液对照组,术后分别依据每组要求经尾静脉给药,共给药7 d,1次/d。于观察期结束时分别经心脏采集外周全血作流式细胞分析CD4+/CD8+。结果:经外源性CORM-2干预后的同种异基因皮肤移植模型的小鼠脾T淋巴细胞亚群(3组)CD4+/CD8+明显低于2组、4组,差异有统计学意义(P<0.05)。结论:外源性一氧化碳释放分子CORM-2对同种异基因皮肤移小鼠淋巴细胞功能有抑制作用。 Objective: Imitation song dynasty style typeface using flow cytometry technique to observe treated with exogenous carbon monoxide CORM-2 same allogeneic skin transplantation in mice model of 7 d receptors in mice spleen lymphocyte subsets CD4^+/CD8^+ change.Method: Taking the SPF ICR mouse, the BALB/C mouse separately as the donor and the acceptor, establishing skin allotransplantation model, and dividing into 4 groups : group 1 of sham-operation group, group 2 of skin allotransplantation group, group 3 of skin allotransplantation +CORM-2 group, group 4 of skin aUotransplantation +iCORM-2 group.Result: T lymphocyte subgroup analysis with FCM indicates that the ratio of mice spleen CD4^+T/CD8^+T in group CORM-2(group 3) was lower than the control group(group 2 and 4) obviously.Conclusion: Exogenous release carbon monoxide molecules CORM-2 on the same allogeneic skin showed inhibitory effect on the function of lymphocyte in mice.
