Detection of HIV/AIDS Viral Loads With Two Different Quantitative Methods

摘要 目的比较2种方法检测人类免疫缺陷病毒(HIV)1型病毒载量的相关性及一致性,为合理评价HIV/AIDS病程进展及抗HIV药物疗效提供参考。方法对四川省凉山州95例在治HIV/AIDS的血浆样本同时用分支DNA杂交实验(bDNA)法和核酸序列扩增实验(NASBA)法检测HIV-1病毒载量,结果进行比较分析。结果该2种方法测得阳性样本均值bDNA法为3.75±1.05 log copies/mL,NASBA法为4.22±1.23 log copies/mL,差异有统计学意义(t=6.09,P<0.05);2种方法测出的病毒载量呈显著直线相关(r=0.852,P<0.01);2种方法测出的病毒载量阳性率差异无统计学意义(χ2=0.836,P>0.05)。结论 bDNA法和NASBA法检测HIV-1病毒载量具有高度的一致性,在实际工作中均可选用,但是两者检测结果的数值不能完全等同。建议抗HIV药物疗效评价最好应用同一种方法的检测结果,使其治疗前后的结果具有可比性和一致性。 Objective To compare the relativity and consistency between two different testing methods for quantitative detection of HIV - 1 plasma viral load, in order to rationally assess the progress of HIV/AIDS and the effieacy of anti - I'tlV drugs. Methods Blood samples of 95 cages of HIV iecton an MDS patients re tested for HIV - 1 viral load by bDA and NASBA. Results The average HIV viral load dt13ted by bDNA and NAS- BA was 3.75 + 1.05 g copies/mL 9/)d 4.22 + 1.23 log copies/mL respectively, and tho difference was a statistically significant betwe-en tl tw.o groups t -: 09, P 〈 0. 05 ). There was a aignificant linear correla- tion between the two methods (r = 0.001 ): The difference showed no statistically significant between the positive rates of the two methods (X 2 =0.836 P〉0. 05). Conclusion There is a high degree coincidence between the two methods for detecting HIV viral load,yet they can not be equally used in the prac- tical work. It is more preferable to employ only one method to evaluat the efficacy of anti - HIV drugs for a a better comparability and consistncy before and after treatment.
