阿科特素体外处理乳腺癌细胞的基因表达通路分析 被引量:1

摘要 近年来有研究表明黑升麻提取物阿科特素具有抗肿瘤活性,但其分子机制仍然不够清楚,研究用基因集富集分析的方法分析了20μg/mL和40μg/mL两种不同质量浓度的阿科特素处理乳腺癌细胞MDA-MB-453 6 h或24 h后的基因表达谱,获取处理后的乳腺癌细胞中的基因集富集情况.结果表明:处理6 h的乳腺癌细胞中没有获得有统计学意义的基因集,而在处理24 h的乳腺癌细胞中与细胞分裂、细胞周期等相关的生物学通路则表达下调,40μg/mL阿科特素处理还可以使得乳腺癌细胞中大分子降解相关通路表达上调.由于阿科特素可以促进细胞凋亡并抑制细胞周期,为其今后用于乳腺癌的预防和治疗的可能性提供了良好的理论依据. Recent studies indicate that the triterpene glycoside actein from the herb black cohosh possess antitumor activity. However, the molecular mechanisms remain unknown. This study seeks to identify change of gene expression profile in human breast cancer cells by treatment with actein. Gene set enrichment analysis was carried out to inspect MDA-MB-453 human breast cancer cells treated with actein at 2 doses, 20 or 40 p.g/mL, for 6 or 24 h. The results showed that gene sets related to cell cycle and cell mitosis were down-regulated in breast cancer cells treated for 24 h, while no macromolecule catabolic significant gene set was enriched in those for 6h. Furthermore, pathways associated with process were up-regulated in breast cells treated with 40 trg/mL for 24h. Since actein activated apoptosis and repressed cell cycle, it may be useful in the prevention and therapy of breast cancer.
