摘要
目的探讨"温阳补气"针法对实验性自身免疫性重症肌无力(EAMG)大鼠血清中转化生长因子(TGF)-β、肿瘤坏死因子(TNF)-α表达水平影响的作用机制和效果。方法采用AChRα1 129-145多肽片段免疫接种雌性Lewis大鼠,建立EAMG大鼠动物模型;随机选取建模成功的EAMG大鼠30只,分成针灸治疗组、药物对照组、模型对照组各10只,并将同期购进未建模的10只大鼠设为空白对照组;针灸治疗组予以"温阳补气"针法治疗,药物对照组予以溴吡斯的明灌胃治疗,其余两组不予任何特殊处置,仅作对照观察;治疗结束后,从四组大鼠尾根静脉采血,采用酶联免疫吸附试验(ELISA)法测定大鼠血清中TGF-β、TNF-α的含量。结果治疗结束后,针刺治疗组EAMG大鼠血清中TNF-α的表达水平显著降低,与模型对照组相比差异显著(P<0.05);TGF-β的表达水平明显升高,与模型对照组相比差异显著(P<0.01)。结论 "温阳补气"针法治疗MG的疗效良好,可通过调节TNF-α和TGF-β的表达水平而发挥治疗MG的作用。
Objective To observe the influence of "warming yang and nourishing qi" acupuncture treatment on the serum expres sion levels of TGF-β and TNF-α of EAMG rats, and analyze the mechanism. Methods AChRal 129-145 peptide fragments was used to im mune Lewis female rats, establish EAMG rat models. The successful EAMG model rats were divided into acupuncture treatment, drug con trol, model control groups, each group had 10 rats; the unmodelled 10 rats which were purchased at the same period were set to blank control group. The acupuncture treatment group was given "warming yang and nourishing qi" acupuncture treatment, the drug control group was giv- en pyridostigmine. After treatment, the serum expression levels of IL-12 and IL-18 were tested by ELISA. Results After treatment, the ser um expression level of TNF-α was significantly reduced, and the level of TGF-β was increased in acupuncture treatment group than that of model control group ( P 〈 0. 05, P 〈 0. 01 ). Conclusions The "warming yang and nourishing qi" acupuncture has good therapeutic effect through adjusting the expression levels of TNF-α and TGFβ.
