摘要
目的构建并繁殖T细胞表达绿色荧光蛋白的LFA-1基因敲除小鼠[Tg(CD4-EGFP);LFA-1-/-]。方法将CD4-EGFP转基因小鼠与LFA-1基因敲除小鼠杂交并将其子代相互杂交,提取小鼠尾部DNA,用PCR法鉴定基因型。根据孟德尔定律,可得到Tg(CD4-EGFP;LFA-1-/-)小鼠。结果 Tg(CD4-EGFP);LFA-1-/-小鼠的构建、饲养繁殖均获得成功。结论成功获得了T细胞表达绿色荧光蛋白的LFA-1基因敲除小鼠。
Objective To construct Tg(CD4-EGFP) ;LFA-1^-/- transgenie mice. Methods CD4-EGFP transgenic mice were crossed with LFA-1 gene mutant mice. The genotype of Tg (CIM-EGFP) ; LFA-1^-/- mice was identified by PCR. Results Tg (CD4-EGFP) ;LFA-1^-/- mice were successfully established. Conclusion The combination of cross breeding and genotype identification is an effective way for acquiring specific transgenic mice.
