4种处理方式对水稻IF分析中染色体形态的影响

Influence on Rice Chromosome Preparation by Four Treatments in IF Analysis

摘要 蛋白免疫荧光(immunofluorescence,IF)分析是确定相关蛋白在染色体上具体位置的重要方法。本研究通过设计4种不同处理方式对水稻单倍体材料T2037的根尖染色体进行IF分析,发现4种处理方式对根尖染色体制备的效果不同。在没有酶解处理的条件下,经磷酸盐缓冲液(phosphate buffer solution,PBS)缓冲液保存的根尖比经过甲醇保存根尖的制片效果好,检测信号强;而酶解后的根尖染色体在两种不同处理方法下,染色体均有较重细胞质背景,检测信号弱。研究表明PBS保存材料且不进行酶解处理的根尖材料更适合于水稻染色体蛋白免疫分析。 Protein Immunofluorescence (IF) analysis is an important method for determining related proteins on chromosome location. This work focused on influence on rice chromosome preparation by four different treatm- ents of root-tip in IF analysis. Without enzyme treatment, the result indicated the cytoplasm background of root- tips, preserved in PBS buffer, was weaker than that preserved in methanol. With enzyme treatment, chromosome morphology was poor in both preservation methods. The result showed that PBS to save material and no enzyme treatment were suitable for rice chromosome IF analysis.
