房颤犬心房间质重塑的相关细胞因子表达 被引量:2

The relationship between atrial fibrillation caused atrial remodeling and expression of related cytokines

摘要 目的研究房颤与心房结构重构的关系以及心房结构重构对心脏间质细胞调控相关的TGFβ1,FGFR-1,VEGF等相关细胞因子表达的影响。方法实验动物随机分为三组:房颤式起搏组、T型钙通道阻滞剂组、正常对照组,24周后取其心房组织进行免疫组化染色,然后使用图像软件测量并比较Ⅰ型胶原和TGFβ1,FGFR-1,VEGF的表达量;分析3组犬心房调控因子与Ⅰ型胶原蛋白表达量的相关性。结果房颤式起搏组的Ⅰ型胶原表达量明显高于T型钙通道阻滞剂组和正常对照组。房颤式起搏组和正常对照组的心房组织TGFβ1表达与Ⅰ型胶原有统计学相关性,而T型钙通道阻滞剂组无统计学相关性;三组心房组织的FGFR-1、VEGF表达量与Ⅰ型胶原均无统计学相关性。结论持续性房颤可能导致心房发生明显的结构重塑,TGFβ1可能具有很强的致心房纤维化作用。 Objective To estimate the relationship between atrial fibrillation (AF) and atrial remodeling, and to study whether atrial strncture remodeling can affact the level of some cytokines, such as TGFβ1, FGFR-I and VEGF, which are closely related with regulation of Myocardial interstitial cells. Methods All the experimental dogs were divided randomly into three groups: 4 dogs got paced with AF-rhythm,5 dogs were treated with T-type calcium channel blockers and the left 5 acted as the no-treatment control group. The atrial tissues were sampled 24 weeks later for Immunohistochemical staining, and then we used imaging analysis software to detect and compare the level of type I collagen, TGF β l, FGFR-1 and VEGF. Furthermore, statistical correlation analysis was made between type I collagen and the 3 cytokines. Results The level of type I collagen was obviously higher in AF-pacing dogs than that in the other two groups. The correlation coefficient between TGF β 1 and type I collagen had high statistical significance in clogs of AF-pacing and the no-treatment control groups. However, there was no statistical correlation between levels of FGFR-I ,VEGF and type I collagen in eaeh of the three groups. Conclusions Persistant AF may result in structure remodeling in atrial, while TGF β 1 may play an important role in atrial fibrosis.
