摘要
目的通过比较四逆汤方药不同组合配伍对大鼠心脏的影响,探讨四逆汤配伍的减毒机制。方法 160只SD大鼠随机分为正常组、生附组、干姜组、炙甘草组、生附+干姜组(3∶3)、生附+炙甘草组(3∶2)、干姜+炙甘草组(3∶2)、生附+干姜+炙甘草组(3∶3∶2),每组20只,各组药材提取液的最终浓度均为1g/ml,根据动物体重按2ml/kg给药。采用十二指肠注射给药后,心电图连续描记120min。以给药后出现频发性室性早搏作为附子心脏毒性标准,按1∶0.8的比例降低/升高给药剂量,每个剂量使用1只动物,以出现心率异常剂量的上1个浓度剂量为开始,引入序贯表,连续观察20只大鼠的心电图变化,依据序贯表中各实验组给药剂量及动物心率异常出现时间,计算各组药物心脏毒性剂量。结果生附组心脏毒性剂量为6.25g/kg,心率异常时间为(12.36±1.15)min,生附+干姜组分别为4.19g/kg、(6.18±1.09)min,与生附组比较,生附+干姜组可显著缩短大鼠心脏毒性发生时间(P<0.05)。其余各组无明显心脏毒性。结论四逆汤中生附片与干姜配伍可增大对心脏的毒性作用,炙甘草可对抗干姜、附子对心脏的毒性作用。
Objective To research the attenuated effect and mechanism of different compositions of Sini Decoction (Counter- flow Cold Decoction) according to their effects on rat hearts. Methods Totally 160 SD rats were randomized into the normal group, Shengfu (Aconiti Radix Lateralis Cruda ) group, Ganjiang (Zingiberis Rhizoma ) group, Gancao (Glycyrrhizae Radix) group, Shengfu + Ganjiang (3 : 3) group, Shengfu + Gancao (3 " 2) group, Ganjiang + Gancao (3 : 2) group and Shengfu + Ganjiang + Gancao (3 : 3 ." 2) group, with 20 in each. The final concentration of medicinal extract in each group was lg/ml and was administrated by injection into the duodenum at 2ml/kg based on animal body weight. The tracings of ECG lasted for 120 minutes. The occurrence of frequent premature ventricular contractions after administration was taken as the cardiac toxicity criteria of Fuzi. The dose was reduced or increased at a ratio of 1 : 0.8 and one rat was used at each dose. The sequential form was introduced according to the former dose when arrhythmia occurred. The changes of ECG in 20 rats were continuously observed. The cardiotoxic dose in each group was calculated according to the sequential form. Results The cardiotoxic dose was 6.25g/kg and arrhythmia time was (12.36±1.15) min in the Shengfu group. The cardiotoxic dose was 4.19 g/kg and arrhythmia time was (6.18 ± 1.09) min in the Shengfu + Ganjiang group. Comparing with the Shengfu group, the occurrence time of cardiac toxicity in the Shengfu + Ganjiang group was significantly shortened (P^0.05). There was no obvious cardiac toxicity in other groups. Conclusion The composition of Shengfupian and Ganjiang in Sini Decoction can increase the cardiac toxicity but Gancao can fight the cardiac toxicity of Ganjiang and Fuzi.
