摘要
目的研究负性协同刺激分子PD-1和CTLA-4表达水平在泡球蚴持续性感染小鼠肝脏中的变化特点,探讨其在泡球蚴感染中的免疫作用。方法将90只雌性Balb/c小鼠随机分为实验组和对照组,实验组通过腹腔接种原头蚴悬液制备泡球蚴感染小鼠模型,对照组腹腔注射等量生理盐水。分别在接种2、8、30、90、180和330 d收集标本。用HE染色观察肝脏病理变化;免疫组化法和实时荧光定量PCR检测PD-1和CTLA-4在肝脏的表达及分布。结果肝脏泡球蚴持续性感染中,PD-1和CTLA-4在感染早期少量表达;感染中晚期表达水平均明显升高,而且均维持一定的高水平。结论 PD-1与CTLA-4表达的增加可能促进泡球蚴感染病情的发展,参与泡球蚴的免疫逃避并利于泡球蚴在机体内的长期寄生。
This study was designed to observe the dynamic change of expression and function of PD-1 and CTLA-4 in liver of Balb/c mice infected persistently with Echinococcus multilocularis (Era). We investigated the function of PD-1 and CTLA-4 and the mechanism of immune escape of Em in mice. Pathological examination, immunohistochemical staining and quantitative real-time PCR (QRT-PCR) detection were used for expression and distribution of PD-1 and CTLA-4. We found that in mice of the experiment groups, sizes of Em cysts and liver impairment gradually enlarged with time in comparison to the mice of the control groups. Suq3risingly, the levels of PD-1 and CTLA-4 expression in liver tissue of the infected mice increased with time, and reached a peak and maintained a high level thereafter. We presumed that increased expressions of PD-1 and CTLA-4 might contribute to the development of alveolar hydatid infection, and be advantageous for alveolar hydatids to evade the inmmne system and live long in vivo.
