摘要
方法:用氯化钾诱发SD大鼠皮层扩散性抑制(CSD),改良 Longa法制作大鼠 MCAO模型,用地高辛标记的 cRNA探针,行原位杂交检测谷氨酸载体EAAC1 mRNA表达。结果:假手术组脑片上单位面积OD为(2 189±263),阳性颗粒数为(43.2±4.7)个。缺血 30min时皮层EAAC1 ,mRNA表达明显上调,经CSD处理后再缺血时,与对照组比较表达水平均明显增高,其中Li 30min时最高。结论:CSD的作用可能与诱导EAAC1合成,增加谷氨酸的摄取有关。
Methods :CSD was induced in rats with 3 mol .L-l KCl,and the modified Longa's method was used to make focal cerebral ischemia model. The expression of glutamate transporter EAACl mRNA were detected by in situ hybridization with DIG-labeled cRNA probe. Results: In sham-operated group, the OD in an area unit was 2l89 ±263,the number of positive cells was 43. 2±1. 7. Following ischemia for 30 min,the expression of EAACl mRNA in cortex was obviously up-regulated. In ex- periment group,the expression of EAACl mRNA was obviously higher than that in control group (P<0. 05). The effect of CSD is maybe inducing the synthesis of EAAC1 and increasing the uptake of glutamate. Conclusion :EAACl may play an im- portant role in the regulation of glutamate concentration following ischemia, the effect of CSD may induce the synthesis of EAAC1 and increase the uptake of glutamate.
