摘要
目的探讨白藜芦醇(Res)对小鼠宫颈癌细胞株U14移植瘤的抑制作用及其相关机制。方法构建615近交系小鼠U14移植瘤动物模型,并随机分为对照组、Res组、顺铂组,每组10只,给予相应药物干预,接种后第26天处死全部小鼠,检测肿瘤体积及重量,计算抑瘤率,通过免疫组织化学染色法检测肿瘤组织微血管密度(MVD),采用荧光实时定量PCR和Western blotting分析肿瘤组织缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)mRNA及蛋白表达水平。结果 Res组和顺铂组抑瘤率分别为50.46%、57.19%;Res组和顺铂组肿瘤体积、肿瘤质量较对照组显著降低(P<0.01);与对照组比较,Res组和顺铂组HIF-1α、VEGF mRNA与蛋白表达水平及MVD明显降低,差异有统计学意义(P<0.01),但Res组与顺铂组比较,差异无统计学意义(P>0.05)。结论 Res通过下调HIF-1α、VEGF表达抑制小鼠宫颈癌的生长。
Objective To explore the inhibitory effect of resveratrol (Res) on transplanted cervix cancer cell U14 in mice, and its related molecular mechanisms.Methods'The 615-strain mice with U14 cervical cancer cells were divided into three groups randomly,namely control group (n=10),Res group (n=lO) and cisplatin group (n=lO).After drug interven- tion,all mice were sacrificed on day 26 posttransplantation.The volumes and weighs of tumors were detected,and tumor inhibition rate was calculated.Microvessel density (MVD) was measured by immunohistochemistry.The levels of hypoxia inducible factor-lc~ (HIF-lc~) as well as vascular endothelia growth factor (VEGF) mRNA and protein expression were measured by real-time quantitative PCR and Western blotting,respectively.Results The tumor inhibition rate was 50.46% and 57.19% in Res group and cisplatin group,respectively.The volumes and weights of tumor was markedly decreased in Res group and cisplatin group compared with control group (P〈0.01).The levels of HIF-Iot as well as VEGF mRNA and protein expression and MVD was significantly lower in Res group than that in control group respec tively (P〈O.O1).However,there was no significant difference in above indicators between Res group and cisplatin group (P〉0.05).Conclusion Res may suppress the growth of cervix cancer by downregulating the expression of HIF-lc~ and VEGF in mice.
