摘要
目的:探讨胰高血糖素样肽-1(GLP-1)在高、低浓度葡萄糖作用下对胰岛β细胞胰岛素原合成的短期影响及随时间的变化趋势。方法:将NIT-1细胞随机分为以下4组:LG组(培养液中含5.6 mmol/L的葡萄糖)、LGG组(培养液中含5.6 mmol/L的葡萄糖和20 nmol/L的GLP-1)、HG组(培养液中含16.7 mmol/L的葡萄糖)、HGG组(培养液中含16.7 mmol/L的葡萄糖和20 nmol/L的GLP-1)。检测基线0 h及葡萄糖和(或)GLP-1刺激后2、4、6、8、10 h各组胰岛β细胞胰岛素原mRNA和蛋白表达。结果:基线时各组胰岛素原表达无统计学差异(P>0.05),刺激2、4、6、8、10 h后HG组、HGG组胰岛素原表达均显著增高(P<0.05),而LG和LGG组无明显变化(P>0.05)。组间比较HGG组胰岛素原表达量显著高于LG组、LGG组及HG组(均P<0.05)。HG组胰岛素原表达量显著高于LG组、LGG组(均P<0.05)。胰岛素原mRNA的相对表达量在刺激后4 h达到高峰,而胰岛素原蛋白的表达量在2 h达到高峰。结论:在高糖条件下,GLP-1短期刺激胰岛β细胞可显著增加胰岛素原合成,GLP-1联合高糖对胰岛素原合成具有协同作用。胰岛β细胞在受到高糖或高糖+GLP-1刺激后胰岛素原蛋白表达的高峰要先于胰岛素原mRNA表达的高峰。
Objective To investigate the short term effect of glucagons-like peptide-1 (GLP-1 on proinsulin biosynthesis by pancreatic β-cells and time-dependent change trend under stimulation of glucose in high and low concentrations. Methods The NIT-1 cells were randomly divided into four groups, LG group (cultured with 5.6 mmol/L glucose), LGG group(cultured with 5.6 mmol/L glucose and 20 nmol/L GLP-1), HG group(cultured with 16.7 mmol/L glucose), HGG group(cultured with 16.7 mmol/L glucose and 20 nmol/L GLP-1). Proinsulin mRNA and protein level at baseline and after 2 h, 4 h, 6 h, 8 h, 10 h stimulation of glucose and/or GLP-1 were assessed by semi-quantitative RT-PCR and HE method respectively. Results At baseline there was no difference among different groups(P 〉 0.05). 2 h, 4 h, 6 h, 8 h, 10 h after stimulation, the expression of proinsulin in HG and HGG groups increased significantly (P 〈 0.05), however LG and LGG groups did not significantly changes (P 〉 0.05). The expression level of proinsulin in HGG group was higher than LG, LGG, HG group (P 〈 0.05). The expression level of proinsulin in HG group was higher than LG, LGG group (P 〈 0.05), Proinsulin mRNA's expression peaked 4 h after stimulation,and proinsulin protein's expression peaked at 2 h. Conclusions Short term GLP-1 stimulation can promote proinsulin biosynthesis and proinsulin gene expression under high concentration of glucose. The combination of GLP-1 and high concentration glucose lead to a synergy effect on proinsulin biosynthesis.
