扩增活化自体淋巴细胞对裸鼠腹膜后脂肪肉瘤移植瘤的抑制作用 被引量:1

摘要 目的 观察扩增活化自体淋巴细胞(EAAL)对裸鼠腹膜后脂肪肉瘤移植瘤生长的抑制作用.方法 提取腹膜后脂肪肉瘤患者外周血单核细胞,以anti-CD3抗体(5 mg/L)和白细胞介素(IL)-2(700 IU/ml)共培养诱导EAAL细胞,流式细胞仪检测细胞表型特征(CD3、CD4、CD8、CD56、CD25).20只裸鼠随机分为低、中、高剂量组和空白对照组,皮下接种人腹膜后脂肪肉瘤细胞2周后,分别经尾静脉输注不同数量的EAAL细胞(3×105、10 × 105和30×105个)和生理盐水,观察肿瘤生长情况.结果 经14d的体外培养后,腹膜后脂肪肉瘤患者EAAL细胞成分中CD8+T细胞、CD3+ CD56+T细胞比例均较前增多,分别为(63.20±15.42)%比(21.85 ±5.77)%、(15.36±7.37)%比(5.23±2.54)%,P< 0.05,而CD4+ CD25+T细胞比例则较前明显减少[(1.78±0.70)%)比(5.39±1.67)%,P<0.05];裸鼠体内实验结果表明,经EAAL细胞治疗后各组肿瘤的体积和重量均较对照组明显缩小,且随着EAAL细胞输注数量的增加,其对肿瘤生长的抑制程度亦逐渐增强.结论 EAAL细胞治疗可有效抑制裸鼠腹膜后脂肪肉瘤移植瘤的生长. Objective To investigate the inhibitory effect of expanded activated autologous lymphocytes (EAAL) against retroperitoneal liposarcoma nude mouse xenograft model.Methods Peripheral blood mononuclear cells (PBMC) were separated from retroperitoneal liposarcoma patients and cultured in anti-CD3 (5 mg/L)-coated flasks with interleukin (IL)-2 (700 IU/ml) to induce EAAL.Flow cytometry was used to analyze the phenotypes of EAAL (CD3,CD4,CD8,CD56,and CD25).Twenty nude mice were randomly divided into 4 groups:low-,medium-and high-dose injection group,in which mice were inoculated subcutaneously with human retroperitoneal liposarcoma cells,and control group.Two weeks after inoculation,different number of EAAL (3 × 105,10 × 105,and 30 × 105) and saline were injected via caudal vein into each group of mice.Meanwhile,tumor volume and weight were measured after treatments.Results During the 14-day culture in vitro,the proportion of CD8 + T cells and CD3 + CD56 + T cells was increased significantly [(63.20 ± 15.42) % vs.(21.85 ± 5.77) %,and (15.36 ± 7.37) % vs.(5.23 ± 2.54)%,P〈0.05,respectively],and by contrast,the population of CD4 + CD25 + T cells was decreased as compared with pre-culture [(1.78 ± 0.70) % vs.(5.39 ± 1.67) %,P 〈 0.05].In human retroperitoneal sarcoma xenograft mice model,EAAL showed a significantly inhibitory effect on the growth of the liposarcoma cells,which resulted in the decrease of tumor volume and weight.Besides,there was dose-response dependent relationship between the inhibitory effect and number of treated EAAL.Conclusion EAAL may potentially serve as an adoptive immunotherapy for human retroperitoneal liposarcoma.
