摘要
建立亲水作用色谱-串联质谱法同时测定乳制品中的纳他霉素和三聚氰胺的方法。利用三氯乙酸溶液-甲醇提取乳制品中的目标成分,正己烷净化,用亲水作用色谱分离,在多反应监测(MRM)模式下以保留时间、离子对(母离子和两个碎片离子)信息比较和峰面积进行定性和定量分析。结果表明,纳他霉素和三聚氰胺在10~200μg/L范围内线性关系良好,定量限(LOQ)分别为135,50μg/kg,方法回收率分别为82.7%~103.1%,85.5%~102.6%,相对标准偏差(n=6)分别为3.2%~5.8%,2.9%~4.9%。此方法可用于乳制品中纳他霉素和三聚氰胺的确证与测定。
A hydrophilic interaction chromatography-tandem mass spectrometry method was developed for simultaneous determination of natamycin and melamine in milk products. Target components were extracted by aqueous solution of trichloroacetic acid and metha nol, purified by n-hexane, separated by hydrophilic interaction chromatography and detected by eleetrospray ionization(ESI) using multiple reaction monitoring (MRM) mode. Qualitative and quantitative analysis was based on the retention time and peak area of the parent ion and two fragment ions. Good linear relationship was achieved in the concentration range of 10 - 200 μg/L for natamycin and melamine. The LOQ levels for natamyein and melamine were 135 μg/kg and 50μg/kg, and recoveries were 82.7%-103.1% and 85.5%- 102.6%, with relative standard deviations(RSDs) of 3.2%-5.8% and 2.9-4.9%, respectively. The method can be used for identification and detection of natamycin and melamine in milk products.
