摘要
目的 观察富血小板血浆(PRP)通过调节细胞周期蛋白p27表达,促进脂肪干细胞(ADSCs)向成骨细胞分化过程中的具体机制.方法 酶消化法获得大鼠ADSCs,观察PRP对大鼠ADSCs向成骨细胞分化的影响.流式细胞仪检测PRP作用与ADSCs对细胞周期和细胞周期蛋白表达的影响,Western blot法检测细胞周期蛋白表达的变化.结果 PRP干预组ADSCs增殖率为41.0%,空白对照组增殖率为7.4%.S期ADSCs的比例由诱导前的26.5%提高到67.9%.结论 PRP通过下调细胞周期抑制蛋白,提高ADSCs的增殖和分化率,促进ADSCs的成骨分化.
Objective To study platelet-rich plasma (PRP) by regulating the expression of Cyclin p27 promote fat stem cells to osteoblast differentiation mechanism.Methods By enzyme digestion rat adipose stem cells,PRP was observed in rat adipose stem cells into osteoblast differentiation.Flow cytometry PRP role and adipose stem cells on cell cycle and cell cycle proteins,Western blotting assay of cell cycle protein expression changes.Results PRP ADSCs intervention group was 41.0% value-added,the blank control group added 7.4%.The proportion of S-phase ADSCs before induction increased from 26.5 percent to 67.9 percent.Conclusion PRP through down the cell cycle inhibitory protein,fat stem cells to improve the added value and differentiation rate,promoting fat stem cells into osteoblasts.
