摘要
以芦丁、十八胺、甲醛为原料,甲醇为溶剂,按照Mannich反应机理合成十八烷基氨甲基芦丁(RuC18)。旨在获得一种适宜的合成方式,既保留芦丁完整的活性结构,同时又增加芦丁脂溶性,合成出具有双亲性的可作为生物治疗功能用的分子。分别通过常规水浴合成和微波合成的方法去探究反应条件对反应速率和产率的影响。得到常规合成的的最佳条件为:单体摩尔比(芦丁:十八胺:甲醛)为1∶2∶12;pH 4.2~4.5;温度75℃,产率最高可达11.2%。微波合成的最佳功率与时间为:150 W,8 min或者200 W,5 min,产率可达18.9%左右,远高于常规的水浴合成方法。并通过FT-IR、1H NMR、13C NMR表征发现,C-取代反应发生在芦丁单元"A"环上的C8位,而且为单取代方式。
According to Mannich reaction mechanism,octodecyl amine methyl rutin( RuC18) was synthesized by using microwave- assisted method and water-bath process,respectively. Rutin,octadecylamine and formaldehyde were used as raw material,and methanol as solvent. The effect of these two synthesis methods on reaction rate and productivity was investigated. In water-bath process,productivity can reach 11. 2%,while mole ratio of rutin,octadecylamine and formaldehyde is 1∶ 2∶ 12,and pH value ranges from 4. 2 to 4. 5,and reaction temperature is 75 ℃. In microwave-assisted synthesis,the productivity is about 18. 9% with the microwave power of 150watts and reaction time of 8 minutes. RuC18 structure is detected by using FT-IR and nuclear magnetic resonance. It was found that the unimolecular substitution reaction occurs in C8position of heterocyclic ring " A".
