摘要
利用超声波辅助皂化法从万寿菊中提取叶黄素,在单因素试验的基础上,对万寿菊叶黄素的超声波辅助皂化工艺进行了探讨,选择超声波功率,皂化温度、氢氧化钠用量和皂化时间4个因素,采用L9(34)正交试验优化提取条件,比色法测定叶黄素含量。结果表明,当超声波功率为500 W,皂化温度为65℃,每克万寿菊颗粒加入4.0 mL 15%氢氧化钠乙醇溶液皂化3 h时,比色法测定万寿菊叶黄素含量最高,为87.34 mg/100 g。各因素对叶黄素含量的影响从大到小依次为超声波功率>皂化温度>氢氧化钠乙醇溶液用量>皂化时间。
In this study,ultrasonic technique was used to assist the extraction of lutein from marigold.Based on the single factor experimental results,four factors,including ultrasonic power,saponification time,saponification temperature and NaOH amount,were chose to optimize the yield maximizing of lutein by using L9 (34) orthogonal test,and the amount of lutein was measured by colorimetric method.The results indicated that when the amount of raw material was 1.00 g,the amount of 15 % Na0H-ethanol solution was 4.0 mL/g,saponification time was 3 hours and extraction temperature was 65.0 ℃,the extraction content of lutein was highest,which was 87.34 mg/100g.The order from large to small which various factors influenced on lutein content was ultrasonic power > saponification temperature > NaOH-ethanol solution amount > saponification time.
