摘要
目的观察氯化两面针碱对斑马鱼胚胎血管生成的影响,并探讨其可能作用机制。方法前期实验基础上设计4个药物浓度组(8.19、10.24、12.80、16.00 mg·L-1),含等量助溶剂DMSO为空白对照组,通过NBT/BCIP血管染色法观察氯化两面针碱对72 hpf斑马鱼胚胎肠下静脉血管发育的影响;半定量RT-PCR法检测血管生成相关基因表达变化。结果氯化两面针碱剂量依赖性抑制斑马鱼胚胎肠下静脉血管生成,下调血管生成相关基因VEGF、VEGFR-2和FGF2的表达。结论氯化两面针碱可以抑制斑马鱼胚胎血管生成,可能机制与影响VEGF、VEGFR-2和FGF2的表达有关。
Aims To investigate the effect of nitidine chloride (NC) on angiogenesis during zebrafish embryos development, and to explore the underlying mechanism. Methods Based on the preliminary experiments, the 24 hpf zebrafish embryos were treated by NC (8.19, 10.24, 12. 80, 16.00 mg· L^-1) and blank control group was treated by 0.5% DMSO. The effects of NC at different concentrations on zebrafish' s subintestinal vessels (SIVs) were observed by NBT/ BCIP vessel staining, and the expression of angiogenesis gene was analyzed by RT-PCR. Results NC dose- dependently showed inhibitory effect on SIVs and down-regulated mRNA expressions of VEGF, VEGFR- 2 and FGF2 in zebrafish embryos. Conclusion NC can inhibit the zebrafish embryos angiogenesis, and the mechanism is probably related to the inhibitory effect on mRNA expressions of VEGF, VEGFR-2 and FGF2.
