摘要
通过花粉管通道法将高粱总 DNA导入春麦甘麦 8号、陇春 1 3号和陇春 1 0号 ,经过多代选择获得了 5个稳定的后代。在高分子量麦谷蛋白亚基分析中 ,甘麦 8号后代 891 44的高分子量麦谷蛋白亚基发生突变 ,较其受体多了 5+1 0亚基 ,而少了 2 +1 2亚基 ;其它几个转基因后代与其受体比较 ,高分子量麦谷蛋白亚基组成未发生变化 ;但是 ,各亚基的相对含量有较大变化。高分子量麦谷蛋白亚基的组成和各亚基含量的变化直接影响小麦品质。本研究对外源总 DNA花粉管通道法导入小麦在改良小麦品质方面的作用进行了讨论。
The total DNA of sorghum was introduced into spring wheat Ganmai No.8,Longchun No.13 and Longchun No.10 by using the pollen tube pathway.Five progenies had been obtained.The studies on storage protein showed that mutation of high molecular weight subunits of glutenin happened in the progeny 89144.Compared with its recipient Ganmai No.8,the high molecular weight subunits 5+10 were presented in 89144 instead of subunits 2+12 of Ganmai No.8.There were no changes on the pattern of HMW subunits of glutenin of other progenies.But the relative contents of high molecular weight subunits of glutenin of the progenies were different from those of their acceptors respectively.Both of the patterns and content of high molecular weight subunits of glutenin affect wheat quality directly.It was discussed that the effect of introduction total DNA via pollen tube pathway might improve wheat quality.
