太原汉族人群17个Y-STR基因座等位基因频率及遗传关系

摘要 目的研究太原汉族人群17个Y-STR等位基因的频率、遗传多态性分布、遗传距离和聚类分析及其在法医学应用研究。方法应用Y-filerTM试剂盒检测102例太原汉族无关男性个体的血样,运用3130 xl遗传分析仪进行基因分型,计算等位基因频率。应用相关分析软件,参考其他地区人群资料,对太原汉族群体的遗传距离和遗传关系进行研究。结果 17个Y-STR基因座共有137个等位基因。基因多样性在0.433 5-0.894 0之间。根据遗传距离分析发现,与公开发表的国内其他群体相关数据资料相比,太原汉族与北京汉族之间的遗传距离最小(0.001 7),与台湾泰雅族人群之间的距离最大(0.469 6);与其他少数民族相比,太原汉族与辽宁满族群体之间的遗传距离最小(0.001 4),与台湾泰雅族人群之间的距离最大(0.4696)。结论 17个Y-STR基因座在太原汉族群体中有较好的遗传多态性,且有较强的个体识别能力及非父排除能力,对建立Y染色体数据库和法医学实际检案都有重要的意义。 Objective To investigate the allele frequency distribution,gene diversity,genetic distance and cluster analysis of 17 Y-specific short tandem repeats (STR) in Taiyuan Han population and to explore their forensic application.Methods Blood samples of 102 unrelated male individuals of Taiyuan Han population were collected to detect 17 Y-STR loci with Y-filerTM system.The 3130xl genetic analyzer was used to analyze the gene typing and calculate the allele frequencies.Cluster analysis and phylogenic trees were applied to investigate the genetic distance among the populations and their genetic relationships.Y-STR data of other populations were collected from publications.Results The 17 Y-STR loci had 137 alleles.The gene diversity(GD) values for each locus ranged from 0.433 5 to 0.894 0.The genetic distance between Taiyuan and Beijing Han populations was the smallest (0.001 7),while the genetic distance between Taiyuan Han population and Taiwan Atayal population was the largest(0.469 6).The genetic distance between Taiyuan Han population and Liaoning Manchu population was the smallest(0.001 4),while the genetic distance between Taiyuan Han population and Taiwan Atayal population was the largest(0.469 6).Conclusion The 17 Y-STR loci reveal high polymorphic genetic distribution,discrimination power(DP) and probability of exclusion (PE),which is very helpful to establish Y-STR database and apply to the forensic cases.
