摘要
目的探讨伊马替尼与顺铂联合对胶质母细胞瘤细胞株U87细胞凋亡及其裸鼠皮下移植瘤的抑制作用,为临床治疗提供实验依据。方法 SubG1法检测U87细胞株在伊马替尼及顺铂单药处理和联合处理时各组细胞的凋亡率;观察不同治疗对U87荷瘤裸鼠移植瘤的抑制作用,绘制肿瘤生长曲线并计算抑瘤率及Q值;测量各组裸鼠体质量变化,评价药物的毒副作用。结果伊马替尼与顺铂联合处理对U87细胞凋亡诱导率显著高于相同剂量的各单药处理组;联合治疗组与相同剂量的各单药治疗组比较,抑瘤率差异有统计学意义(P<0.05),Q值为1.264;与顺铂高剂量组比较,抑瘤率差异无统计学意义(P>0.05)。结论伊马替尼与顺铂联用有协同效应,二者联用可降低顺铂使用剂量,减轻对裸鼠的毒副作用。
Objective To investigate the inhibitory effects of imatinib combined with cisplatin on human glioblastoma cell line U87 and xenograft in nude mice,and provide experimental evidence for clinical application.Methods U87 cell lines were treated with imatinib and/or cisplatin,and the cell apoptosis ratio was accessed by SubG1 measured with FACS.The effects on different treatment groups of nude mice xenografts were observed,tumor growth curve was drewand the tumor-inhibition rate and Q value were calculated.To measure the nude mice weight of each group,and evaluate side effects of drugs.Results Apoptosis ratio treated with imatinib combined cisplatin was significantly higher than the same dose single drug group respectively.There was significant difference in tumor-inhibition rate between combined group and the same dose single drug group（P0.05）,and the Q value was 1.264,there was no significant difference between combined treatment group and high dose cisplatin group（P0.05）.Conclusion There is a synergistic effect between imatinib and cisplatin,combination of the two drugs can reduce the dose of cisplatin and mitigate the toxic side effects on nude mice.
