摘要
以"神舟八号"搭载的苜蓿种子为材料,经过筛选获得高株、多分枝等变异材料,利用HPLC、MSAP等方法对紫花苜蓿DNA甲基化状况进行了研究。结果表明,与地面对照相比,空间搭载苜蓿的DNA甲基化水平提高3%~8%,且增加水平因变异类型而异,其中多分枝变异的材料DNA甲基化水平最高,搭载后无明显表型变异的DNA甲基化水平最低。此外,DNA甲基化状态也发生显著变化,搭载材料表现为甲基化和去甲基化两种变化,其中以超甲基化为主。
To uncover the effect of space flight factors on the DNA methylation of alfalfa will be help- ful to expand knowledge on the epigenetic mechanism of mutation. Based on the methods of HPLC and MSAP, the percentage of cytosine DNA methylation was analyzed and compared the ground controls. Data showed that the percentage of cytosine DNA methylation increased by 3 % -8% after the space flight. And the degree of increase varied with the types of mutation. The mutation with increased plant height and branches had the highest percentage of cytosine DNA methylation. Moreover, there were two types of cy- tosine DNA methyaltion change. Conclusion is that the space flight factors can increase the level and status of cytosine DNA methyaliton significantly which provide the primary proof for the epigenetic mechanism of mutation and will be helpful to understand the molecular mechanism of mutations induced by space flight factors.
