高产辅酶Q_(10)光合细菌的筛选及鉴定 被引量:2

摘要 对池塘污泥中富集分离的5株光合细菌产生的辅酶Q10(CoQ10)进行定性定量分析,筛出CoQ10产量较高的菌株P1,并对其进行系统鉴定。菌株P1为革兰氏阴性茵,杆状,菌体大小为(0.5~0.8)mm×(1.0~1.9)μm;适合光照厌氧培养,适宜生长温度为28~36℃,适宜pH为7.0~7.5;菌体含有细菌叶绿素a和类胡萝卜素;多种有机化合物均可作为光合作用的电子供体和碳源,蛋白胨和硫酸铵是其生长的较好氮源。16SrDNA序列系统发育分析表明,菌株P1在系统进化树上与GenBank序列号为AB167544的沼泽红假单胞菌菌株聚为一类,初步确定菌株P1为沼泽红假单胞菌。沼泽红假单胞菌P1菌株CoQ10产量较高,在19mg/L左右,至少能稳定传代10次。 Five photosynthetic bacteria producing coenzyme Q10 ( CoQ10 ) were enriched and isolated from pond sludge and qualitative and quantitative analyses on the CoQ10 were carried out. One strain named P1 with higher CoQ10 yield was isolated and identified systematically. The strain P1 was gram - negative and rod - shaped of(0.5~0.8)mm×(1.0~1.9)μm. It could not grow aerobically in dark, and the optimal temperature and pH value for growth were 28 - 36℃ and 7.0 - 7.5 respectively. Its thallus contained bacteri- ochlorophyll a and carotenoids. Various organic compounds could be used by strain P1 as photosynthetic elec-tron donors and carbon sources. Peptone and (NH4)2SO4 were better nitrogen source for its growth. Phyloge-netic analysis based on 16S rDNA sequence revealed that P1 was clustered into one group with Rhodopseudo-monas palustris, whose accession number in GenBank was AB167544. It preliminarily showed that P1 was Rhodopseudomonas palustris. The CoQ10 yield of strain P1 was higher, about 19 mg/L, and could be subcul-tured stably at least 10 generations.
