摘要
目的观察癫持续状态(SE)后脑水肿及血脑屏障破坏的动态变化。方法建立氯化锂-毛果芸香碱大鼠SE模型,成模后0、4、24和72 h分别采用干湿重法测脑含水量,伊文思蓝检测血脑屏障通透性,电镜观察血脑屏障超微结构。结果 SE成模后24 h组脑组织含水量显著增加(P<0.01),伊文思蓝荧光强度显著增高(P<0.01),电镜显示各组血脑屏障基本完整。结论 SE后血脑屏障通透性和脑水肿程度均增加,以成模后24 h最为显著。
Aim To observe the dynamic changes of brain edema and damage of blood-brain barrier(BBB) associated with status epilepticus in rats. Methods The status epilepticus(SE) model was induced by the administration of lithium chloride and pilocarpine. At 1 hour after achieved Racine level 4, rats were treated with i.p. diazepam. The rats were divided into five groups as follows: 0 h, 4 h, 24 h, 72 h after the termination of SE, and a control group. Thirteen rats were in each group. The water content in brain with a dry-wetweighing technique were measured. The severity of brain edema were evaluated by using Evans blue albumin extravasation and fluorescence intensity measurement. The structures of blood-brain barrier were observed by electron microscope. Results Compared with the control group, the water content of brain were higher in the SE groups, significantly in the group of 24 h after SE(P<0.01), and which was higher than the other SE groups, but without statistically significant difference(P>0.05). Fluorescence intensity of Evans blue albumin was observed at 24 h after the termination of SE, compared with the other groups, increased statistically significantly(P<0.01). By using electron microscope, the structures of BBB were basically normal and the morphologies of tight junction and endothelia cell were found almost complete in all groups. Conclusion The increased permeability of blood-brain barrier and the severity of brain edema associated with status epilepticus were observed, with the most severity occurred at 24 h after seizure. There were no significant abnormalities in the morphology of vascular endothelial cell.
