摘要
目的:研究复方葛花制剂对酒精性肝损伤小鼠的干预作用。方法:将50只小鼠分为空白对照组10只,造模组40只。空白对照组(A组)以普通饲料喂养,自由饮水;造模组随机分为模型组即阳性对照组(B组),复方葛花低剂量组(C组)、复方葛花中剂量组(D组)、复方葛花高剂量组(E组)、每组10只,每天固定时间(上午9:00),给予造模组小鼠52%(V/V)红星二锅头白酒灌胃,12 mL/kg,每天1次,A组代以等量蒸馏水。2 min后,C组、D组和E组分别以0.2 g/mL、1 g/mL和5 g/mL的中药浓缩液灌胃,10 mL/kg;B组以等量蒸馏水灌胃。连续灌胃28d,检测小鼠血清TG、TC、HDL-C、LDL-C及MDA、SOD、GSH-PX水平及肝组织病理学改变。结果:模型组的血清TC、TG水平较其余各组显著升高(P<0.05),有统计学意义;血HDL-C水平模型组较空白组降低(P<0.05);血LDL-C五组相比无统计学差异(P﹥0.05)。与空白对照组相比,模型组小鼠MDA水平升高(P<0.05),复方葛花高剂量组MDA含量与模型组相比降低(P<0.05);复方葛花低剂量组(C组)、复方葛花中剂量组(D组)、复方葛花高剂量组(E组)SOD活性比模型组相比升高(P<0.05);而GSH-PX活性在各组间比较无显著性差异(P>0.05)。结论:复方葛花制剂能改善小鼠脂代谢并降低机体氧化应激水平,从而改善酒精性肝损伤。
Objective: To study the intervention effect of Compound Gehua Preparation on alcoholic liver damage of mice. Meth- ods : 50 mice were divided into control group with 10 mice ( group A) and molding group with 40 mice. Group A were given nor- mal diet and could drink water freely; molding group were randomly divided into model group or positive control group (group B), Compound Gehua Preparation group with low dosage (group C), Compound Gehua Preparation group with moderate dosage (group D), Compound Gehua Preparation group with high dosage (group E), 10 mice in each group. At the fixed time in each morning (9 o'clock), the mice of molding group were given gastric perfusion of Red Star Erguotou of 52% ( V/V), 12 mL/kg, once per day. Group A were administrated distilled water of equal mnount. After 2 min, Group C, D and E were given Chinese medical concentrated solutions of 0.2 g/mL, g/ml and 5g/m respectively; Group B was given gastric perfusion of distilled water of equal amount. The intragastric administration lasted for 28 days, and the levels of TG, TC, HDL - C, LDL - C, MDA, SOD and GSH - PX in the mice's serum as well as the pathological changes of liver were detected. Results : The levels of TC and TG in the serum of the group B increased significantly( P 〈 0.05 ), which was of statistical significance. Compared with group A, group B presented increased level of MDA (P 〈 0.05 ). The content of MDA in group E decreased when compared with group B ( P 〈0.05). The activity of SOD in group C, group D and group E increased when compared with group B(P 〈0.05). Howev- er, the activity of GSH - PX among different groups had no significant difference ( P 〉 0.05 ). Conclusion : Compound Gehua Preparation can better the lipid metabolism in mice and lower the level of oxidative stress of the body, thus improving the alcohol- ic liver damage.
