摘要
从陕西省关中地区患多发性浆膜炎和关节炎猪场的32个样品中分离到6株细菌,并对其进行了形态观察、菌落纯培养、生化特性鉴定、PCR检测等,最终确定分离菌株均为副猪嗜血杆菌。该结果表明,关中地区存在副猪嗜血杆菌病流行;对副猪嗜血杆菌分离株最适培养基进行了筛选,将分离菌株分别接种于LB、TSA、巧克力琼脂等固体培养基上,接种12h后通过观察菌落大小,筛选出最适合副猪嗜血杆菌生长的固体培养基TSA(加0.1g/L NAD和50mL/L的犊牛血清)和固体培养基巧克力琼脂(加0.8g/L NAD)。同时,将副猪嗜血杆菌分离菌株接种于LB、TSB等液体培养基中,培养至16h后,每隔4h分别取样,通过与麦氏比浊管对比确定菌液中的菌数,筛选出最佳液体培养基为TSB(加0.1g/L NAD和50mL/L的犊牛血清)。
To certify whether the Glasser's disease existed in Guanzhong District,Shaanxi province,6 bacte- ria were isolated from the swine which were characterized by polyserositis and fibrinopurrlent polyarthritis. Then the bacteria were identified based on the cultural characteristics,morphologic characteristics, biochemical characteristics ; The findings also suggested that Haemophilus parasuis widely existed in this area, providing certain experimental basis for studies of Haemophilus parasuis disease in this area. We screened the optimum medium of Haernophilus parasuis, the Haemophilus parasuis isolates were inoculated on LB,TSA, chocolate agar solid medium, to observe the colony size by 12 h after inoculation, selec- tion of optimum Haemophilus parasuis growth of TSA and chocolate agar solid medium. At the same time, the Haemophilus parasuis isolates were inoculated on LB, TSB and other liquid culture medium, training to 16 h, every 4 h respectively through sampling, and with Michael's opacity compared to determine the bacterial liquid about bacteria, screening out the best liquid medium TSB.
