摘要
目的观察洛铂对胃癌细胞SGC-7901细胞增殖的影响,并探讨其作用机制。方法将对数生长期的SGC-7901细胞分阴性对照组(不加药物)和实验组(加入不同浓度洛铂),采用倒置显微镜观察不同浓度下细胞生长状态;MTT法观察对细胞的增殖抑制;Western印迹方法分析不同浓度洛铂对SGC-7901细胞内Bcl-2蛋白表达水平的影响。结果倒置显微镜下观察,洛铂能抑制细胞增殖,出现明显的凋亡学形态特征。MTT法显示洛铂能抑制细胞增殖,并呈浓度及时间依赖关系。Western印迹结果显示洛铂能使SGC-7901细胞Bcl-2蛋白表达水平下降。结论洛铂能明显抑制胃癌SGC-7901细胞增殖,诱导凋亡,可能与调节SGC-7901细胞Bcl-2蛋白表达水平相关。
Objective To observe the effect of lobaplatin on the proliferation of gastric cancer cell SGC-7901 and to study the mechanism.Methods SGC-7901 cells in the logarithmic phase were divided into negative control group(without drugs) and experimental group(adding different concentrations of lobaplatin).The inverted microscope was used to observe cell growth at different concentrations.MTT method was used to observe the inhibition of the gastric cancer cell proliferation.Western blot was used to detect the expression of Bcl-2 in SGC-7901 cells that were treated by different concentrations of lobaplatin.Results Lobaplatin inhibited cell proliferation in a concentration-and time-dependencty manner.Western blot results showed that Bcl-2 protein level decreased after lobaplatin treatment.Conclusion Lobaplatin could significantly inhibit gastric cancer SGC-7901 cell proliferation and induce apoptosis,possibly related to the regulation of Bcl-2 protein expression.
